Quantifying Lipid Accumulation in Adipocytes

Lipid accumulation was detected by Oil Red O staining on days 4 and 8 of differentiation program. The adipocytes were washed twice with PBS and then fixed for 30 min with 4% paraformaldehyde (Biosharp, Hefei, China). The cells were rinsed twice with PBS, stained with 0.2% (m/v) Oil Red O (Sigma-Aldrich) dissolved in isopropanol (Damao Chemical Reagent Factory, Tianjin, China) for 30 min at 37°C. The lipid droplets were evaluated by the Imager A2 fluorescence microscope (Carl Zeiss, Werk Gottingen, Germany) and representative figures were shown. To determine lipid content in the differentiated adipocytes on days 4 and 8, isopropanol (0.5ml/well of a 12-well plate) was added to the plates as reported previously (33 (link), 34 (link)). After the extracted dye was removed via pipetting to a new 96-well plate (200 µl/well), and the optical density was detected spectrophotometrically at 510 nM with the Synergy H4 Hybird microplate reader (BioTek, Winooski, VT, USA).

Mtb H37Rv Infection of THP-1 Macrophages

Mtb standard strain H37Rv (ATCC 27294) was preserved by the laboratory. THP-1 human macrophages were purchased from the Shanghai Institute of Cell Research, Chinese Academy of Sciences. Phorbol-12-myristate-13-acetate (PMA) and Ficoll lymphocyte separation medium were purchased from Sigma (USA). Synthesis of Luminescent g-C3N4 Coating

Because the fluorescence emission wavelength of long afterglow phosphor needed to match the absorption wavelength of g-C3N4 and has good water resistance, we chose sky-blue long afterglow phosphor (Sr2MgSi2O7:(Eu2+, Dy3+), SLAP) produced by Dalian Luminous Technology Co. Since the specific surface area of g-C3N4 prepared from urea was larger, urea (AR, 98%) produced by Fuchen Chemical Reagent Co., Ltd. (Tianjin, China) was purchased as a precursor for the preparation of g-C3N4. Ten thousand molecular weight polydimethylsiloxane (PDMS) was selected as the film-forming substance for the preparation of the coating and purchased from Shandong Dayi Chemical Co. (Shandong, China). Isopropanol (AR, ≥99.7%), anhydrous ethanol (AR, ≥99.7%), and ethyl orthosilicate (TEOS, AR, 98%) were obtained from Tianjin Damao Chemical Reagent Factory (Tianjin, China). Pyrogenic silica (AS-150) was obtained from Shenyang Chemical Co. Anti-settling agent DeuRheo202P was obtained from Hemmings Deqian Chemical Co. (Changzhou, China). The organic solvents xylene (AR, 99%) and methyl isobutyl ketone (MIBK, AR, 99%) were purchased from Tianjin Comio Chemical Reagent Co. The catalyst bismuth neodecanoate (DY-20) was purchased from Shanghai Deyin Chemical Co. (Shanghai, China), and the bismuth content was 20 ± 0.5%.

PLGA Nanoparticles for Drug Delivery

Male rabbit (2.0 kg) was obtained from the Laboratory Animal Unit of Kunming Medical University (Kunming, China). Animal was anesthetized with pentobarbitone sodium (200 mg/kg,i.p.). Then drew blood from rabbit pericardium to heparinized tubes. All experiments about animals performed in this study were approved by the Committee on the Use of Live Animals in Teaching and Research of Yunnan University of Nationalities.
