ELISA kits for Insulin-like growth factor I (IGF1, #ab211651, intra-assay coefficient of variation (CV) 2.3%) and Insulin-like growth factor-binding protein 3 (IGFBP3, #ab211652, intra-assay CV 3.1%) were purchased from Abcam (Cambridge, MA), and ELISA kit for Insulin-like growth factor-binding protein 2 (IGFBP2, #DGB200, intra-assay CV 5%) was from R&D Systems (Minneapolis, MN), and ELISA kit for Anti-mullerian hormone (AMH, #AL-124, intra-assay CV 3.7%) was purchased from AnshLabs (Webster, TX). The plasma samples from each time point of selected subjects were diluted with dilution factors of 1:80, 1:100, and 1:2000 for IGF1, IGFBP2, and IGFBP3 detection, respectively; and for AMH measurement, dilution factors of 1:10 and 1:500 were used for samples from females and males, respectively. All ELISA measurements were conducted according to manufacturer’s protocol. Duplicated wells were used to average the absorbance values and the concentration of target proteins in the plasma were calculated based on the calibration curve (R2 = 0.9848, 0.9999, 0.9936 and 0.9971 for IGF1, IGFBP2, IGFBP3, and AMH, respectively.) and dilution factor after subtraction of background.

Serum Biomarker Quantification

Blood samples were obtained on the day following hospitalization and were centrifuged at 3,500 rpm for 10 min to separate the serum, which were stored at −80°C. GH was measured using an enzyme-linked immunosorbent assay (ELISA) with an ELISA detection kit (ab190811, Abcam). IGF-1, IGF-2, IGF receptor-1, and IGF receptor-2 levels in prospective cohort were detected using an immunoassay (QAH-IGF-1, RayBiotech). IGF-1 level in cross-sectional cohort was detected using an ELISA detection kit (ab211651, Abcam).

Serum Myokine and Growth Factor Profiling

Check if the same lab product or an alternative is used in the 5 most similar protocols
Resting blood samples were collected early in the morning for fasting specimens and at least 48 h post any exercise. The collected blood samples were processed to serum and stored at −80 °C until serum myokine analysis. Serum myokine levels for irisin, IL-6, IL-15, SPARC, OSM, and myostatin, were analysed using multiplex magnetic bead panels (HMYOMAG-56k-15 Huma, Millipore, Billerica, MA, USA), and serum decorin (ab99998, Decorin Human ELISA Kit, Abcam, Cambridge, United Kingdom), IGF-1 (ab211651, Human IGF-1 SimpleStep ELISA Kit, Abcam, Cambridge, United Kingdom), and IGFBP-3 (ab211652, Human IGFBP3 SimpleStep ELISA Kit, Abcam, Cambridge, United Kingdom) levels were analysed using appropriate enzyme-linked immunosorbent assay (ELISA) kits.

Quantification of VEGF and IGF-1 in Conditioned Media

Check if the same lab product or an alternative is used in the 5 most similar protocols
Conditioned medium from 2D and 3D-cultured cells was stored at -80 • C until analysis. Unconditioned complete medium was used as a negative control. Human-specific ELISA plates were obtained from Abcam for VEGF (ab222510) and IGF-1 (ab211651). Assays were carried out following the instructions provided with each kit. Culture media was diluted to within the assay linear range if necessary.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
