Fluorescence-activated cell sorting (FACS) analyses of sub-G1 fractions using propidium iodide staining for DNA fragmentation. Briefly, MPT-S1 cells were harvested in 15 mL tubes, fixed with 70% ethanol, washed twice with PBS and stained with propidium iodide (PI) (50 μg/ml) and RNase A (100 μg/ml) for 30 min at 37 °C. At least 10,000 events were analyzed by flow cytometry (Coulter EPICS Elite ESP; Beckman Coulter, Fullerton, CA, USA) with the excitation set at 488 nm and emission at 610 nm. Data were analyzed using Flowing Software version 2.5.1 (Turku Bioscience, Turku, Finland).
