Multicolor Imaging of Pb-Stressed Bacteria

The bacteria cells were cultured in 100 mL LB medium for 24 h in the presence of 20 mg/L of Pb (II) and collected by centrifuge at 8000 × g for 10 min, followed by suspension in saline buffer. RNA FISH and Immunofluorescence Dual Labeling

Cell samples plated on coverslips precoated with poly-l-lysine (SIGMA) were fixed in 4% PFA and then processed for RNA FISH and immunofluorescence as described previously (Naganuma et al. 2012 (link)). Double fluorescence TSA was carried out using FITC-labeled doRPS12 and DIG-labeled doSRSF1 RNA probes detected with HRP-labeled anti-fluorescein and Cy3-conjugated Tyramide or HRP-labeled anti-digoxigenin antibodies with Cy5-conjugated Tyramide (PerkinElmer). After probe hybridization and washing, the TSA procedure was performed in accordance with the manufacturer's protocol, adding a 30-min treatment with 1% of H2O2 between two TSAs to quench residual peroxidase activity from the first TSA reaction. Confocal images were acquired under a FLUOVIEW FV1000 confocal laser scanning microscope (Olympus) and a Nikon Confocal system A1Rsi (Nikon) equipped with a Plan Apo VC 60× objective lens (NA 1.40, Nikon).
