Mouse Strain Generation and Maintenance

C57BL/6 and B6.SJL mice were purchased from Taconic Biosciences (Germantown, NY). A breeding pair of Rag2−/−IL-2Rγ−/− mice was purchased from Taconic Biosciences, and these mice were subsequently bred in-house. Rorc.cre and R26R-EYFP mice were purchased from The Jackson Laboratory (Bar Harbor, ME). Rorc.cre mice were bred with R26R-EYFP mice in-house to produce RORγt fate mapping mice. NFIL3−/− mice were a generous gift from Dr. Hugh JM Brady (15 (link)). NFIL3+/+, NFIL3+/−, and NFIL3−/− mice were subsequently bred in-house. All mice were maintained in pathogen free facilities at Brown University. Mice of both sexes were included and no differences were observed.

Murine Immune Cell Lineage Evaluation

IL-12Rβ2fl/fl mice were generated at the Brown University Transgenic Facility. Both age and sex-matched mice (6 − 14 weeks) were used for this study. All experimental procedures were approved by the Institutional Animal Care and Use Committee (IACUC) of Brown University, and were conducted in accordance with institutional guidelines for animal care. All mice were maintained at Brown University in AAALAC-accredited, pathogen-free facilities.

Transgenic Mouse Strains for Immunology

In this study, female and male IL-4Rα/IL-5-deficient BALB/c, Rag2tm1FwaII2rgtm1WjlRAG2IL-2Rγ-deficient BALB/c and C57BL/6 (RAG2IL-2Rγ−/−), and NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ (NOD scid gamma, NSG) BALB/c mice were used. These mouse strains were provided by the Institute of Medical Microbiology, Immunology and Parasitology (IMMIP), University Hospital Bonn (UKB) in Germany. RAG2IL-2Rγ−/− mice were purchased from Taconic Biosciences Inc (Cologne, Germany) and the other two strains were bred at the IMMIP and UKB animal facilitates, but originally, IL-4Rα/IL-5−/− were a gift from Prof. Dr. Klaus Matthaei (Matthaei, Stem Cell & Gene Targeting Laboratory, ANU College of Medicine, Biology and Environment, Canberra, Australia) and NSG mice were purchased from Jackson Laboratory (Bar Harbor, Maine, USA). Mice were kept under SPF conditions following German animal protection laws and EU guidelines 2010/63/E4 and had access to food and water ad libitum. Mice were shipped in filter topped boxes to the Research Foundation for Tropical Diseases and the Environment (REFOTDE), Buea, Cameroon in agreement with the veterinary office in Bonn, Germany. Upon arrival, mice were housed in the animal facility of the Research Foundation for Tropical Diseases and the Environment, University of Buea under SPF conditions following Cameroonian animal protection laws.

Murine Model of Immune Deficiency

Rag2-/- and Rag2-/-Il2rγ-/- mice, on a C57Bl/6 background, were originally obtained from Taconic Biosciences (Troy, New York, USA). Male and female mice between 6 and 10 weeks of age, an average weight of 25 g, were used in this study. All mice were bred and housed in specific pathogen-free rooms within the Tulane University School of Medicine animal facilities, in accordance with the guidelines of the institution.
