GC-MS Analysis of Acrylamide and Acetamide

From Merck Co., the reagents including potassium hydroxide, potassium hexaferrocyanide, acetic acid, methanol ethanol (analytical grade), hydrochloric acid (HCl) 37 %, dipotassium hydrogen phosphate (K2HPO4), acetamide (purity ≥ 99 %) (GC grade), acrylamide (purity ≥ 99.9 %), tetrachloroethylene (to conduct GC analysis), potassium hexaferrocyanide (carrez I) and zinc acetate (carrez II), were obtained. From Sigma-Aldrich Co., acetamide (purity ≥ 99 %) and Xanthydrol, were purchased. Agilent 7890A gas chromatographic (Agilent Co., USA) device coupled to an Agilent 5975c Mass selective detector (MSD inert); the column was an Agilent capillary column HP-5 ms (5 % phenyl siloxane/ 95 % methyl polyorganosiloxane; length × I.D.: 30 m × 0.25 mm; df: 0.25 µm).
For preparation of carrez solution I, 10.6 g of potassium hexaferrocyanide was dissolved in distilled water (100 mL). Carrez solution II was prepared by mixing 21.9 g of zinc acetate with acetic acid (3 mL), then was adjusted the volume to 100 mL with distilled water. The primary standard solution of acrylamide and acetamide (2000 μg/mL) was prepared in methanol. To obtain a working solution, the upper standard solution was diluted with methanol. Stock and working solutions were stored at 4 °C.

Ultrastructural Analysis of Collagen Fibrils

Dorsal skin samples were collected from 7 months old WT and Sc65KO animals (n = 4) and fixed overnight at 4°C in 2.5% glutaraldehyde (Electron Microscopy Sciences (EMS), Hatfield, PA, USA), 0.05% malachite green (Sigma Aldrich) in 0.1M sodium cacodylate buffer, pH 7.2 (EMS). After washing with 0.1M sodium cacodylate buffer, the samples were post-fixed for 2 hrs with 1% osmium tetroxide (EMS), 0.8% potassium hexaferrocyanide (Sigma Aldrich) for 2 hours and 1% tannic acid (EMS) for 20 min. The samples were rinsed with molecular grade water and stained with 0.5% uranyl acetate (EMS) for 1 hour then dehydrated with a graded alcohol series and propylene oxide before embedding in Araldite/Embed 812 (EMS). Sections (50nm) were cut on a Leica UC7 ultra-microtome and collected on formvar carbon coated slot grids and post-stained with uranyl acetate and lead citrate. Imaging was done using a Technai F20 (FEI, Netherlands) at 80kV. Analyses of collagen fibril diameter (>200 fibrils/mouse) and of collagen inter-fibril space were performed with the Leica Application Suite v 3.0 image analysis software (Leica Microsystems, Milan, Italy) on 5 sections for each mouse (n = 3) at the magnification 19000X.

Spinal Cord Ultrastructure Imaging

Spinal cord ventral roots were dissected and fixed overnight at 4 °C in 2.5%
glutaraldehyde (Electron Microscopy Sciences)/0.05% malachite green (Sigma) in 0.1M sodium
cacodylate buffer, ph 7.2 (EMS). After washing with 0.1M sodium cacodylate buffer, the
samples were postfixed for 2 hours with 1% osmium tetroxide (EMS)/0.8% potassium
hexaferrocyanide (Sigma) for 2 hours and 1% tannic acid (EMS) for 20 min. The samples were
rinsed with molecular grade water, stained with 0.5% uranyl acetate (EMS) for 1 hour, and
then dehydrated with a graded alcohol series and propylene oxide before embedding in
Araldite/Embed 812 (EMS). Thin sections were cut on a Leica UC7 ultramicrotome, collected
on formvar carbon coated slot grids, and post stained with uranyl acetate and lead
citrate. Imaging was taken with a Technai F20 (FEI) at 80kv.

Biogenic Amines Analysis in Non-Alcoholic Beer

Hydrochloric acid (37%), 1-octanol, acetonitrile, sodium chloride, acetone, potassium hydroxide (KOH), ethanol, methanol, 2-nitrophenyl octyle ether (NPOE), dipotassium hydrogen phosphate (K2HPO4), potassium hexaferrocyanide and zinc acetate were purchased from Merck (Darmstadt, Germany). Isobutyl chloroformate was obtained from Sigma-Aldrich (St. Louis, MO, USA). Ten non-alcoholic beer samples were randomly purchased from different supermarkets (Tehran, Iran).
