Immunohistochemical Analysis of Lung Tissue

Check if the same lab product or an alternative is used in the 5 most similar protocols
Lung tissue samples were fixed in 10% formalin and processed for paraffin embedding. Blocks were sectioned and stained with antibodies for specific target antigens: αIL-10 (Acris, PM1121, Herford, Germany), αIL-10R (Abcam, ab197666, Cambridge, UK), αCD3 (Zytomed, RBK024, Berlin, Germany), αPD1 (Zytomed, 516-18632) and αTTF1 (Dako, M3575, Hamburg, Germany). Stained slides were scanned and digital hole slide images were performed using a digital slide scanner (Scan 150, 3D Histech Ltd, Budapest, Hungary) at the task group on digital pathology at the Institute of Pathology, Friedrich-Alexander-Universität Erlangen-Nürnberg (FAU), Erlangen, Germany. Hole slide images were visualised by the CaseViewer (Version 2.0, 3D Histech Ltd). The immune reactive score (IRS) or the area fraction quantification was analysed with ImageJ software Version 1.46 (Bethesda, MD, USA), open source by Wayne Rasband. The detailed quantification method is described in the supplements.

Characterization of Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
For each newly established cell line, cultured tumor cells were centrifuged, fixed, and embedded into paraffin cell blocks according to routine cytology procedures. The cultured cells and xenografts were characterized for their morphological characteristics by routine hematoxylin and eosin stain, mucin content, and immunohistochemistry with a panel of monoclonal antibodies against cytokeratins (CK7 [M7018; DAKO {Agilent Technologies, Santa Clara, CA, USA}], CK20 [M7019; DAKO]) and the thyroid transcription factor TTF-1 (M3575; DAKO). All cell lines were subjected to DNA fingerprinting with markers STR D11S922, D11S4099, and D11S4152.

Evaluating TTF-1 Expression by IHC

Check if the same lab product or an alternative is used in the 5 most similar protocols
For the evaluation of TTF-1 expression by immunohistochemistry (IHC), a mouse MoAb against TTF-1 (M3575; Dako©, Glostrup, Denmark) was used on a Benchmark XT platform (Ventana-Roche, Tucson, AZ, USA). When tumor cells presented nuclear staining, the case was considered TTF-1-positive. Data indicating the intensity, distribution and percentage of positive cells were also collected.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
