Annexin V Apoptosis Assay for M231 Cells

Briefly, 100,000 cells (M231 control cells or NR4A1-null cells) collected from the fresh culture (cell confluency no more than 80%) were first washed with PBS and resuspended in 1x binding buffer, 5 μl Annexin V-PE probe together with DAPI (1:5,000 diluted) were added into the cell suspension and cells were incubated for 5–10 minutes before flow cytometry analysis. Dead cells or apoptotic cells were stained positive for both Annexin V-PE and DAPI.
