For weight loss experiments, 6- to 8-week old female WT C57Bl/6N mice were treated with 100μg of anti-IL-12p80 antibody (Rush University, clone a3-1d) or with Armenian Hamster polyclonal IgG isotype control (Biolegend; #400940) intraperitoneally one day prior to 100 EID50 H5N1-VN/PR8 challenge and 3 days following infection. Mice were monitored for weight loss as described above. For BAL experiments, 6- to 8-week old female mice were treated with 100μg of anti-IL-12p80 antibody or with Armenian Hamster polyclonal IgG isotype control (Biolegend; #400940) intraperitoneally one day prior to 100 EID50 H5N1-VN/PR8 challenge and sacrificed for BAL at 3 dpi as described above. Additionally, recombinant IL-12p80 (Biolegend; 573102) was intranasally administered to 6- to 8-week old male C57Bl/6N mice one day post-infection with H5N1-VN/PR8. 50ng of IL-12p80 was diluted in 50uL of PBS/0.01% BSA with WT control mice receiving 50uL of PBS/0.01%. Mice were sacrificed for BAL at 3 dpi as described above.
