Hepatocyte Cell Lines and Knockout

The LO-2 human normal hepatic cell line and the QGY-7701 and HUH7 HCC cell lines were obtained from ATCC. The HepG2, Bel-7404, SK-Hep-1, and Hep3B HCC cell lines were obtained from the National Collection of Authenticated Cell Cultures (NCAC) in China. Primary human hepatocytes (HH, mixed donor) were obtained from Lonza (HUCS10P). Cell lines were cultured with Eagle's minimum essential medium (EMEM, Gibco) supplemented with 10% fetal bovine serum (FBS, Gibco). HH cells were cultured in HCM™ Hepatocyte Culture Medium BulletKit™ (CC-3198, Lonza). All cells were incubated at 37°C and 5% CO2. hsa-miR-582-5p knockout (miRKO) cell lines were generated by GenePharma with the CRISPR/Cas9 system.

Establishing Primary Dermal Fibroblast Cultures

Primary dermal fibroblast cultures were established from skin biopsies from patients F1-II:1 and F2-II:2 and two unaffected and unrelated individuals (WT1 and WT2), following standard procedures77 . Primary dermal fibroblast cell lines, HEK293T cells (ATCC, catalog no. CRL-3216, RRID:CVCL_0063) and LX-2 cells (Millipore, catalog no. SCC064, RRID: CVCL_5792, gift from H. Weiping (Duke-NUS Medical School, Singapore)) were cultured in complete High Glucose Dulbecco’s Modified Eagle Medium (HyClone) supplemented with 10% fetal bovine serum (FBS) (Biological Industries) and 2 mM L-glutamine (Biological Industries). Hc3716-hTERT cells (a gift from P. Kaldis (Institute of Molecular and Cell Biology, A*STAR, Singapore)) were cultured in HCM Hepatocyte Culture Medium BulletKit (Lonza, catalog no. CC-3198) supplemented with 5% FBS and 10% human serum (Sigma-Aldrich, catalog no. H4522). All cell lines were maintained in a humidified atmosphere at 5% CO2 and 37 °C and tested negative for mycoplasm using the MycoAlert Mycoplasma Detection Kit (Lonza, catalog no. LT07-118).

Cell Culture Maintenance Protocol

HepG2/C3A cells were obtained from the American Type Culture Collection (ATCC, CRL-10741) and maintained in Eagle's Minimum Essential Medium (ATCC) with 10% FBS (HyClone). HepaRG 5F parental cells were purchased from Sigma-Aldrich and maintained with the HCM Hepatocyte Culture Medium BulletKit (Lonza). Cells were incubated in a humidified atmosphere at 37°C with 5% CO2 and routinely verified to be mycoplasma free by using the MycoProbe Mycoplasma Detection Kit (R&D Systems). Cell counts were obtained with a Countess II Automated Cell Counter using trypan blue staining.
