LCMV Infection in Genetically Modified Mice

IL-4Rα KO BALB/cByJ mice were generated by backcrossing IL-4Rα KO BALB/cJ (The Jackson Laboratory) with BALB/cByJ mice for at least five generations. All animal experiments were done with approved Institutional Animal Care and Use Committee protocols at the University of Minnesota. LCMV Armstrong was provided by D. Mouse Strain Acquisition and Maintenance

C57BL/6J (stock number: 000664), BALB/cByJ (stock number: 000651), RAG2 deficient (C.B6(Cg)-Rag2tm1.1Cgn/J) (stock number: 008448), RAG2-, IL-2Rg-deficient (C;129S4-Rag2tm1.1Flv Il2rgtm1.1Flv/J) (stock number: 014593) were purchased from the Jackson Laboratory (Bar Harbor, Maine). BALB/c IL-5 deficient mice (11 generations derived from IL-5 deficient C57BL/6), and BALB/c IL-13 deficient mice was obtained from A. McKenzie (MRC Laboratory of Molecular Biology, Cambridge, UK)64 (link),65 (link). All mice were bred in our animal facility at the Keck School of Medicine, University of Southern California (USC). Mice were maintained at macroenvironmental temperature of 21–22 °C, humidity (48–52%), in a conventional 12:12 light/dark cycle with lights on at 6:00 a.m. and off at 6:00 p.m. In all, 4–8 weeks-old age and sex matched male mice were used in the studies. All animal studies were approved by the USC institutional Animal Care and Use Committee and conducted in accordance with the USC Department of Animal Resources’ guidelines.

In Vivo BALB/c Mouse Infections

For in vivo infections, 5-week-old female BALB/c ByJ (Jackson Laboratory, Bar Harbor, USA) or 5-week-old female immunoglobulin heavy chain-knockout BALB/c mice (C.Cg-Igh-Jtm1Dhu; Taconic Biosciences, Germantown, USA) were used. At appropriate endpoints, mice were euthanized by intraperitoneal injection of tribromoethanol in tert-amyl alcohol at a 1:40 dilution in PBS (2.5% Avertin). Mice were kept under specific pathogen-free conditions at the Louisiana State University Department of Laboratory Animal Medicine. All mouse protocols were approved by the Institutional Animal Care and Use Committee at Louisiana State University.

Genetically Modified Mouse Models

Mouse Strains for PTR Studies

Mouse models for immunological studies

Aged Naive CD4+ T Cell Transfer

Naive CD4+ T cells were purified from young (5–8 wk old) and aged (14–18 months old) HNT.Thy1.1/Thy1.2 mice on a BALB/c background. The HNT TcR recognizes amino acids 126–138 (HNTNGVTAACSHE) of PR8 HA (41 (link)).
The mice were bred and housed in specific pathogen-free conditions at UMass Medical School. Recipients of cell transfers include 8–10 wk old DO11.10 TcR Tg mice that were obtained from Taconic Biosciences (Germantown, NY), CD4 KO or intact BALB/cByJ mice bred at UMass from breeders obtained from Jackson Laboratory (Bar Harbor, ME). BMDC were derived from either BALB/c ByJ or from Il6−/− BALB/c ByJ, both obtained from Jackson Laboratory. All experimental animal procedures were conducted in accordance with the University of Massachusetts Medical School Animal Care and Use Committee guidelines and those from NIH.

Mouse Diversity Panel Protocol

This panel of isogenic mice was chosen based on priority strains from the Mouse Diversity Panel.26 (link) Four mice were used per strain. Male mice were housed four per cage in polycarbonate cages on a 12-hour light/dark cycle (lights on at 7 am), with access to food and water ad libitum. All procedures were approved by the Institutional Animal Care and Use Committee and followed the guidelines set forth by the National Institutes of Health (NIH) Guide for the Care and Use of Laboratory Animals.
