Vybrant proliferation kit

After evaluating pharmacodynamic interactions, we examined the effect of the synergistic drug combination of GDC and ADA on cell proliferation time-dependently. In a 96-well plate, the cells were seeded (3 × 10³ cells/well) and treated with ADA or GDC alone or in combination at concentrations below the IC₅₀ values. After 24–72 h of incubation, according to the manufacturer’s instructions, the proliferation of cells was determined using the Vybrant Proliferation Kit (Thermo Fisher Scientific, United States). GraphPad Prism 8.4.3 and a two-way ANOVA were employed for statistical analysis, followed by a Tukey test for multiple comparisons.

Cells were plated at 3 x 10³ cells/well in a 96-well plate and treated with ADA or DOX alone or combined at a concentration below the IC₅₀. The proliferation of cells was determined after 24–72 hrs of incubation, using the Vybrant Proliferation Kit (Cat no. V-13154, Thermo Fisher Scientific USA).
