Immunohistochemical Analysis of Lung Tissue

Lung tissue samples were fixed in 10% formalin and processed for paraffin embedding. Blocks were sectioned and stained with antibodies for specific target antigens: αIL-10 (Acris, PM1121, Herford, Germany), αIL-10R (Abcam, ab197666, Cambridge, UK), αCD3 (Zytomed, RBK024, Berlin, Germany), αPD1 (Zytomed, 516-18632) and αTTF1 (Dako, M3575, Hamburg, Germany). Stained slides were scanned and digital hole slide images were performed using a digital slide scanner (Scan 150, 3D Histech Ltd, Budapest, Hungary) at the task group on digital pathology at the Institute of Pathology, Friedrich-Alexander-Universität Erlangen-Nürnberg (FAU), Erlangen, Germany. Hole slide images were visualised by the CaseViewer (Version 2.0, 3D Histech Ltd). The immune reactive score (IRS) or the area fraction quantification was analysed with ImageJ software Version 1.46 (Bethesda, MD, USA), open source by Wayne Rasband. Paraffin-embedded Lung Tissue Analysis

Lung lobes were removed, fixed in 10% formalin-PBS solution, dehydrated, and embedded in paraffin. Five-micrometer-thick lung sections from paraffin blocks were stained with hematoxylin and eosin for visualization of lung tumors. Stained slides were then scanned using a digital slide scanner (Scan 150, 3D Histech Ltd). Slide images were analyzed with the CaseViewer software (Version 2.0, 3D Histech Ltd). Hematoxylin and eosin staining were performed in collaboration with the Institute of Pathology at the University Hospital Erlangen.

Quantifying Murine Lung Tumor Burden

Representative parts of the sectioned mouse lung were fixed in 10% formalin-PBS solution, dehydrated, and embedded in paraffin. Five-micrometer-thick lung sections cut from paraffin blocks were stained with hematoxylin and eosin for visualization of lung tumors. Stained slides were scanned using a digital slide scanner (Scan 150, 3D Histech Ltd) and slide images were visualized using the CaseViewer software (Version 2.0, 3D Histech Ltd). Staining and scanning of slides was performed in cooperation with the Institute of Pathology in Erlangen
To analyze the tumor load in the lung for each mouse, the lung sections were histologically evaluated after Hematoxylin and Eosin staining. A pathologist analysed in a blinded codified manner the tumor load of the murine lungs. Moreover, each section was analyzed by means of area [µm²] and number of foci. The sections were investigated under the microscope with an object magnified 10 times (Carl Zeiss, AxioCam MRC) and live pictures were directly transmitted to the Zen 2 blue edition software (Carl Zeiss Microscopy GmbH). With the help of the graphic tool, the foci were localized, circled and automatically measured by means of area. The obtained data corresponding to each mouse was used for comparisons as well as correlation analyzes and transferred into graphs (GraphPad Prism 8).
