Catechol's Effect on COX Enzymes

The direct effect of catechol on COX-1 and COX-2 enzymatic activities was measured using the COX inhibitor screening assay according to the manufacturer's instructions [6] (link). In brief, catechol (final 1–50 µM) was incubated with COX-1 or COX-2 in a reaction buffer for 10 min. Thereafter, AA was added as a substrate and incubated for an additional 2 min. This was followed by the addition of 0.1 N HCl and saturated stannous fluoride solution to stop the reaction. COX-1- or COX-2-mediated PGE2 production was then measured using the Cayman ELISA kits.
