Lipid Nanoparticle Synthesis and Characterization

Tetrachloroauric acid (HAuCl4·3H2O) (520918) and sodium citrate tribasic trihydrate (S4641) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Cell culture media RPMI 1640 (10-040-CV) was obtained from Corning Inc. (Corning, NY, USA). FBS (16000-044) and Penn-Strep (15140-122) were purchased from Life Technologies (Grand Island, NY, USA), Opti-MEM was from Thermo Fisher Scientific (Waltham, MA, USA). The scrambled control siRNA (cat. SIC001), siRNAs against human MICU1 (SASI_Hs01_00070249), CBS (SASI_Hs01_00214623), and CAV1 (SASI_Hs01_00199504) were procured from Sigma-Aldrich. The following primary antibodies were purchased from the specified vendor: rabbit monoclonal anti-MICU1 (#12524, Cell Signaling Technology, Danvers, MA, USA), rabbit polyclonal anti-CBS (#sc-67154, Santa Cruz Biotechnology, Dallas, TX, USA), anti-CAV1 (#SAB871521112, Sigma Aldrich), anti–glyceraldehyde-3-phosphate dehydrogenase (GAPDH) (Sigma-Aldrich), and anti-Ki67 (no. ab833, Abcam).

Western Blot Protein Analysis

Samples were run on a 10% polyacrylamide SDS-PAGE gel, transferred onto nitrocellulose membrane, and blocked for 30 min with 5% milk in TBS containing 0.1% Tween-20 (TBS-T). Membranes were probed with one of the following primary antibodies: 1:1000 anti-CLTC (Sigma, C1860), 1:1000 anti-CAV1 (Sigma, C4490) 1:1000 anti-Rac1 (ThermoFisher, PA1-091), 1:3000 anti-actin (abcam, ab6226), or 1:1000 anti-EGFR (Cell Signaling Tech, 4267). For standard experiments, membranes were washed with TBS-T, probed with goat anti-rabbit or goat anti-mouse HRP conjugated antibodies (Pierce), incubated with ECL substrate (Pierce) according to manufacturer’s instructions, and exposed to film. For quantitative experiments, membranes were probed with goat anti-mouse or donkey anti-goat IR Dye conjugated antibodies (Licor). Membranes were washed with TBS and signal was detected using a Licor Odyssey system.
