Apoptosis Analysis of Liver Cancer Cells

Apoptosis was determined by fluorescence activated cell sorting (FACS) analysis using the Annexin V‐FITC detection kit (KeyGEN BioTECH, Nanjing, China) as described by the manufacturer. HUH‐7, 7721 and HEPG2 cell lines were plated in six‐well tissue plates (4 × 105 cells/well) and incubated overnight. Proliferating cells were treated with or without bazedoxifene for 12 hours. Cells were trypsinized and centrifuged at 720 g for 5 minutes. After washing twice with PBS, the cells were then harvested and incubated with fluorescein isothiocyanate‐conjugated Annexin V and propidium iodide dye (PI) following the manufacturer's protocol before evaluation by flow cytometry (FACS Caliber; BD Biosciences Franklin Lakes, NJ, USA). CellQuest software was used to analyze apoptosis.
