Fabrication of Ring-Shaped Micropatterns

Ring-shaped micropatterns were fabricated as previously described15 (link),29 (link). Briefly, a master mold was generated on a silicon wafer with the SU-8 3025 photoresist (Micro Chem) and chromium masks with ring-shaped features (500 μm outer diameter and 200 μm inner diameter). To fabricate the polydimethylsiloxane (PDMS) stamps, the prepolymer and curing agent (Dow Corning) were mixed at a 10:1 ratio and baked at 60°C for 3 hours after being cast onto the mold. Titanium-gold coated glass slides were printed with 1-octadecanethiol (C18; Sigma-Aldrich), using the PDMS stamps, and subsequently incubated with non-adhesive [HS-(CH2)11-EG3] (EG3; ProChimia) overnight. The slides were coated with 50 μg/mL fibronectin (Sigma-Aldrich) for 30 minutes prior to cell seeding.
