Adenosine Receptor Ligand Preparation

Cyclopentyladenosine (CPA), 5’-(N-ethylcarboxamido)adenosine (NECA), and 8-Cyclopentyl-1,3-dipropylxanthine (DPCPX) were purchased from Sigma-Aldrich, Munich, Germany. 2-[4-(2-carboxyethyl)phenethylamino]-5′-N-ethylcarboxamidoadenosine (CGS-21680), 1-deoxy-1-(6-[([3-lodophenyl]methyl)-amino]-9H-purin-9-yl)-N-methyl-β-D-ribofuranuronamide (IB-MECA) and N-(4-Acetylphenyl)-2-[4-(2,3,6,7-tetrahydro-2,6-dioxo-1, 3-dipropyl-1H-purin-8-yl)phenoxy]acetamide (MRS 1706) were obtained from Tocris (Distributed by BIOZOL Diagnostica, Eching, Germany). Stock solutions (5 mM up to 100mM) were prepared in Ethanol (CPA) or DMSO (all other) and stored at –20°C. Further dilutions were carried out in distilled water.

Characterization of A3 Adenosine Receptor

Origene Technologies (Rockville, MD, USA). DOTAP Liposomal Transfection Reagent
was purchased from Sigma Aldrich (Steinheim, Germany), and the anti-dNGFR
antibody was purchased from Chromaprobe (Maryland Heights, MO, USA). Reference
agonists 2-Cl-IB-MECA, IB-MECA, CGS21680 and NECA were purchased from Tocris
Bioscience (Bio-techne, Abingdon, UK). All other chemicals and reagents used
were purchased from the same suppliers as described previously [18 (link)] or synthesized at NIDDK, National Institutes of
Health as reported [19 (link)–33 (link)].

Evaluating Analgesic Effects of A3AR and H4R Agonists

The selective A3AR agonist, IB-MECA (0.5–1 mg/kg; Tocris Bioscience, Milan, Italy), and the selective H4R agonist, VUF 8430 (10–30 mg/kg; Tocris Bioscience, Milan, Italy) were dissolved in sterile saline solution and intraperitoneally (i.p.) administered on day 8 after CCI surgery. Control animals received an equal volume of vehicle. Behavioral measurements were performed before and 30 min, 1 h, 2 h, 3 h and 5 h after compounds injection.

Fluorescent Binding Assay for A2A Receptor

CGS 21680 hydrochloride, IB-MECA, SCH 442416 and PSB-10 hydrochloride were purchased from Tocris Bioscience (Bristol, UK). Forskolin was purchased from Sigma-Aldrich (St. Louis, MO, USA). For non-radioactive binding assays, tag-lite buffer (TLB) was obtained from Cisbio Bioassays (LABMED, Codolet, France). The Tb derivative of O6-benzylguanine is commercialized as HaloTag-Lumi4-Tb (SHALOTBC, Cisbio Bioassays, LABMED, Codolet, France). A2AR antagonist (SCH 442416 derivate) conjugated to a red fluorescent probe was purchased from Cisbio Bioassays (L0058RED, LABMED, Codolet, France). The intellectual property of TLB components and of fluorophore structure is owned by Cysbio Bioassays.

Effects of Sunitinib and IB-MECA on HL60 Cells

The HL60 cell line were obtained from European Collection of Cell Culture (England). RPMI 1640 medium and MTT was purchased from Sigma Aldrich (USA) and the medium supplements L-Glutamine, HEPES and antibiotics mix (100 U/ml penicillin and 100 µg/ml of streptomycin) were from Invitrogen (UK), while foetal bovine serum (FBS) was from Biosera (UK). Antibodies anti-PKCα (phospho T497) pPKCα and total PKC were from Abcam (UK), while glyceraldehyde 3-phosphate dehydrogenase (GAPDH) (14C10) rabbit mAb and anti-rabbit linked IgG antibody conjugated to horseradish peroxidase were from Cell Signalling (UK). RNAlater, Ambion mirVana miRNA Isolation Kit, Applied Biosystems MicroRNA Reverse Transcription Kit, TaqMan Universal PCR Master Mix II (uracil N-Glycosylase not included), Applied Biosystems primers assays (U6, hsa-miR155, hsa-miR-15a, hsa-miR-16-1, rno-miR-1, hsa-miR-27a, hsa-miR-133a and hsa-miR-133b) were purchased from Life Technologies (USA).
