Sphere Formation Assay for CTCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
CTCs and primary tumor cells were trypsinized and suspended in serum-free Neurobasal-A medium as single-cell suspensions, followed by seeding into 6-well plates at 5,000 cells per well. The cells were treated with or without 1 μM XAV939. After incubation for 8 days, the spheres were imaged and counted under an Axiovert 50 microscope (Zeiss) equipped with an AxioCam HRM CCD camera (Zeiss).

Limiting Dilution Assay for CTCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
In vitro limiting dilution assay was performed as previously described(18 (link)). Briefly, CTCs and primary tumor cells were trypsinized and suspended in serum-free Neurobasal-A medium as single-cell suspensions, followed by seeding into 96-well plates at 1, 5, 10, 20. 40, 80 cells/well. The cells were treated with or without 10 μM XAV939. After incubation for 10 days, the occurrence of spheres was observed under an Axiovert 50 microscope (Zeiss) and the wells with at least one sphere was recorded as positive.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
