Comprehensive HIV Biomarker Assessment

A fasting venous blood sample was collected and analyzed for total cholesterol, LDL-cholesterol, HDL-cholesterol, HbA1c, and glucose[35 (link),37 (link)]. CD4+ T-cell count (cells/uL) was measured as a routine analysis by flow cytometry as previously described[35 (link)]. HIV viral load (copies/mL) was measured with a polymerase chain reaction (PCR) quantitative kit (COBAS®AmpliPrep/COBAS®TaqMan® 48 System; Roche, Basel, Switzerland) according to the manufacturer’s instructions. Nadir CD4 was recorded as the lowest CD4+ T-cell count measured during HIV-infection[35 (link)]. As previously described, plasma concentrations of interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α), were measured by Meso-Scale Discovery MULTI-SPOT plate (MSD, Gaithersburg, MD)[35 (link)], and T-cell subsets (percentage of activated (CD38+HLA-DR+) and senescent (CD28-CD57+) CD8+ T-cells) were determined using a six-colour FACSCanto flow cytometer (Becton Dickinson, Franklin Lakes, NJ)[35 (link)].

Quantitative HBV-DNA PCR Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Quantitative HBV - DNA real - time PCR tests were completed using a COBAS AmpliPrep / COBAS Taqman 48 system (Roche, Branchburg, NJ, USA). Viral nucleic acids were extracted from 500 µl patient serum with a COBAS AmpliPrep automated extractor according to the manufacturer’s instructions. The measurement interval of the kit for HBV- DNA is 20 - 1.7 x 108 IU/ml.

Hepatitis B Biomarker Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
Serum HBsAg and HBeAg levels were detected by an automated electrochemiluminescence immunoassay using a Roche Cobas e601 (Roche Diagnostics, Basel, Switzerland). The upper limit of the HBsAg concentration is 5200 IU/mL. The HBV DNA load was determined using the completely automated COBAS AmpliPrep-COBAS TaqMan 48 system (v.2.0; Roche Molecular Systems, Branchburg, NJ, USA) with a lower detection limit of 20 IU/mL. The linear determination range was 2.0 x 10-1.7 x 10 8 IU/mL. The PreS1-Ag levels were determined by a standard enzyme-linked immunosorbent assay (Alpha, Shanghai, China) according to the manufacturer instructions.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
