Multispectral Imaging and Spatial Analysis

Slides were imaged using the Vectra 3.0 slide scanner and visualized in Phenochart whole slide viewer (Perkin Elmer). Twenty multispectral images per each tumor biopsy were acquired using the 20X objective (200X absolute magnification), with core biopsies, and specimens with less than 20 fields of view or a peritumoral area of less than 350,000 µm2 excluded from the analyses (n = 24/85). Image analysis was performed using the Inform quantitative pathology software (Perkin Elmer) to identify cells, their spatial location and the expression of the above markers on a cell by cell basis. Cell phenotypes were assigned using the quantitative pathology module of TIBCO® Spotfire® 6.0.0 software.

Multispectral Tissue Imaging Protocol

Tissue sections that underwent single-plex fluorescent staining for each fluorophore were imaged using the Vectra 3.0 slide scanner (Perkin Elmer) under the appropriate fluorescent filters (cy3 for Opal570, Texas Red for Opal620, cy5 for Opal650 and Opal690, FITC for Opal520, and DAPI) in order to produce the spectral library required for multispectral analysis. A whole slide scan of the multiplex tissue sections produced multispectral fluorescent images visualized in Phenochart, from which a minimum of 3 regions of tumor and peritumor representative of the entire section were selected where possible for imaging at 20x power for further analysis.
