The levels of triglyceride (TG), blood urea nitrogen (BUN), total billiubin (T-bill), glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in plasma were enzymatically measured using commercially available kits (ARKRAY, Inc., Minami-ku, Kyoto, Japan). Plasma total cholesterol, low density lipoprotein (LDL)-cholesterol and HDL-cholesterol were determined using HDL and LDL/very low density lipoprotein (VLDL) Assay kit (E2HL-100, BioAssay Systems, Hayward, CA, USA). The plasma concentration ofleptin and insulin were measured based on the ELISA method using commercial rat leptin and insulin ELISA kit (Leptin Rat ELISA ab100773, Abcam, Cambridge, MA, USA; Insulin, 80-INSRT-E01, ALPCO, Cambridge, MA, USA).

Metabolic Biomarkers in Rat Models

Check if the same lab product or an alternative is used in the 5 most similar protocols
The fasting glucose and insulin levels were assessed using commercially available kits formulated specifically for rats (#90010, Crystal Chem, Elk Grove Village, IL, USA and #DIGL-100, BioAssay Systems, Hayward, CA, USA) following each supplier’s instructions. In addition, we calculated the values from the homeostatic model assessment of β-cell function (HOMA-β) for each animal using the previously published equation [30 (link)]: HOMA-β = (Fasting insulin (ng/mL) × 20)/(Fasting glucose (mg/dL) − 3.5). Levels of total cholesterol (CHOL) and triglyceride (TG) serum levels were determined using multi-enzyme-based kits (#MBS726298, MyBioSource, San Diego, CA, USA, and #ECCH-100, BioAssay Systems, Hayward, CA, USA, respectively). Serum and hepatic levels of high-/low-density lipoprotein cholesterol (LDL-c/HDL-c were assayed using a special colorimetric kit (#E2HL-100 BioAssay Systems, CA, USA). Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase were analyzed using assay kits (#EALT-100 and EASTR-100, BioAssay Systems, CA, USA). All procedures were performed according to the manufacturer’s instructions for each kit (n = 8 samples/group).

After blood was drawn by cardiac puncture, plasma was isolated using Microvette CB 300 LH tubes (Sarstedt). The concentrations of total cholesterol, LDL/VLDL cholesterol, HDL cholesterol and triglycerides were determined using enzymatic colorimetric assays (#E2HL-100 and #ETGA-200; BioAssay Systems) as per the manufacturer's instructions. Glucose levels were detected using the Contour®Next EZ Blood Glucose Monitoring System with Contour®Next test strips.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
