SmartPool siRNA from Dharmacon. The Amaxa Nucleofector and Cell Line
Nucleofector kit T (Lonza Cologne GmbH) were utilized according to
the manufacturer’s protocol. Cells were transfected with 0.25–0.5
μg of siRNA per 1 million cells and plated in regular growth
medium. The cultures were allowed to rest for 24 h before experiments
were performed and the efficiency of knockdown was determined via
immunoblot analyses.

The following siRNAs (FlexiTube siRNA) were purchased from Dharmacon: PTP4A2 (SMART Pool, M-009078-01-0005, GE Healthcare Europe GmbH, Dharmacon), NOTCH1 (SMART Pool, L-007771-00-0005, GE Healthcare Europe GmbH, Dharmacon), and negative control (SMART Pool, Non-Targeting siRNA#1, GE Healthcare Europe GmbH, Dharmacon). Transfection was performed using lipofectamine RNAimax kit (Thermofisher Scientific, 13778075) in accordance with the manufacturer’s instructions, with siRNA at a final concentration of 10 nM in Opti-MEM I (Thermofisher scientific, 31985070). Cells were harvested 48 h after transfection.
