Hematological Profiles in Animal Study

Check if the same lab product or an alternative is used in the 5 most similar protocols
All animals were fasted for 4 hr before necropsy and blood collection. The blood samples were collected from the abdominal vena cava using a syringe with a 26-gauge needle under isoflurane anesthesia (Ilsung Pharm, Seoul, Korea) and collected into vacutainers containing EDTA-3K (Becton Dickinson, Franklin Lakes, NJ, USA). The samples were analyzed within 20 min of collection using an automated hematology analyzer (Hemavet 950, Drew Scientific, Waterbury, CT, USA). The following parameters were determined in this study: total erythrocyte counts (RBC), hemoglobin concentration (HGB), hematocrit (HCT), mean cell volume (MCV), mean cell hemoglobin (MCH), mean cell hemoglobin concentration (MCHC), platelet count (PLT), whole leukocyte counts (WBC), neutrophils (NEU, %), eosinophils (EOS, %), basophils (BASO, %), lymphocytes (LYM, %) and monocytes (MONO, %).

Five male and five female rats in each group were fasted overnight before the necropsy and blood collection. Blood samples were taken from the abdominal aorta using a syringe with a 21-gauge needle under isoflurane anesthesia (Ilsung Pharm, Seoul, Korea) and collected into vacutainers containing EDTA-3K (Becton Dickinson, Franklin Lakes, NJ, USA). The samples were analyzed within 20 min of collection using an automated hematology analyzer (F-820, Sysmex, Kobe, Japan) The following parameters were determined in this study: total erythrocyte counts, hemoglobin concentration, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration, platelet count, and whole leukocyte counts.

Comprehensive Blood Analysis Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
All animals were fasted overnight before necropsy and blood collection. Blood samples were taken from the abdominal aorta using a syringe with a 24-gauge needle under Isoflurane anesthesia (Ilsung Pharm, Seoul, Korea) and were collected into vacutainers containing EDTA-3K (Becton Dickinson, Franklin Lakes, NJ, USA). Samples were analyzed within 20 min of collection using an automated hematology analyzer (Hemavet 950, Drew Scientific, Waterbury, CT, USA) and an automated coagulation analyzer (HumaClot Duo, Human, Wiesbaden, Germany) with plasma obtained after blood was mixed with 3.2% sodium citrate (9NC sodium citrate, Becton Dickinson). The following parameters were determined in this study: total erythrocyte counts (RBC), hemoglobin concentration (HGB), hematocrit (HCT), mean cell volume (MCV), mean cell hemoglobin (MCH), mean cell hemoglobin concentration (MCHC), red cell distribution (RDW), platelet count (PLT), mean platelet volume (MPV), prothrombin time (PT) and activated partial thromboplastin time (APTT), whole leukocyte counts (WBC), neutrophils (NEU, %), eosinophils (EOS, %), basophils (BASO, %), lymphocytes (LYM, %) and monocytes (MONO, %).

Each blood sample was transferred to CBC bottles (EDTA 3 K; BD Biosciences, San Jose, CA, USA) and assayed on a hematology analyzer (Advia 120E; Siemens Healthcare, Malvern, PA USA). Hematological measurements included eosinophil (EOS), mean corpuscular hemoglobin concentration (MCHC), hematocrit (HCT), neutrophil (NEU), hemoglobin (HGB), mean corpuscular volume (MCV), large unstained cell (LUC), lymphocyte (LYM), red blood cell (RBC), white blood cell (WBC), platelet (PLT), mean corpuscular hemoglobin (MCH), monocyte (MONO), mean platelet volume (MPV), basophil (BASO), red cell distribution width (RDW), and hemoglobin distribution width (HDW).

Hematological Analysis in Fasted Rats

Check if the same lab product or an alternative is used in the 5 most similar protocols
The rats were fasted overnight before necropsy. Blood was collected from the abdominal aorta under anesthesia with isoflurane. Three milliliters of blood was collected in a CBC bottle (EDTA 3 K; BD Biosciences, San Jose, CA, USA) and analyzed using an autohematoanalyzer (Advia120E; Siemens). The remaining blood was transferred into Vacutainer® tubes (sodium citrate 3.2%, BD Biosciences) and centrifuged at 3,000 rpm for 10 minutes. The plasma was separated and clotting time was determined using a coagulometer (ACL 7000; Instrumentation Laboratory, Bedford, MA, USA). The hematological parameters studied were as follows: total leukocyte and differential leukocyte (neutrophil, lymphocyte, monocyte, eosinophil, and basophil) count, total erythrocyte count, hemoglobin concentration, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration, reticulocytes, platelets, prothrombin time, and activated partial thromboplastin time.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
