Monocyte Activation by Bacterial Lipids

Fatty acid standards C14:0, C14:0-3OH, NaOH, HCl and tert-butyl methyl ether were purchased from Sigma (Zwijndrecht, The Netherlands). Methanol and n-Hexane were from JT Baker and n-Hexane from Biosolve.

GmbH (Kandel, Germany). Polybutylene terephthalate (PBT, Mw ∼ 16,150 Da). ULC-MS-grade methanol (MeOH), ethanol
(EtOH), and n-hexane were acquired from Biosolve
B.V. (Valkenswaard, The Netherlands). Tetrahydrofuran (THF) and hexafluoroisopropanol
(HFIP, ≥99% purity) were supplied by Sigma-Aldrich Chemie B.V.
(Zwijndrecht, The Netherlands). All organic solvents
were used without further purification. 2,5-Dihydroxybenzoic acid
(DHB, 98% purity), dithranol (DT, ≥90% purity), 2′,4′,6′-trihydroxyacetophenone
monohydrate (THAP, ≥99.5% purity), silver trifluoroacetate
salt (AgTFA, 98% purity), and trifluoroacetic acid (TFA, ≥99%
purity) were purchased from Sigma-Aldrich.

Antioxidant and Fatty Acid Analysis Protocol

Standards of epicatechin, hyperoside, and ideain were purchased from Extrasynthese. All the chemicals were of analytical grade.

Lipid Extraction and Derivatization Protocol

Acetonitrile, n-hexane, methanol, 2-propanol, and water were purchased from Biosolve BV (Valkenswaard, The Netherlands). Chloroform was from Rathburn Chemicals (Walkerburn, UK). Ammonium formate was from Sigma-Aldrich (Buchs, Switzerland). All solvents and chemicals were HPLC grade. Sulfuric acid and butylated hydroxytoluene (BHT) were from Sigma-Aldrich (Steinheim, Germany), 18O-labeled water (i.e., H218O) was from Medical Isotopes (Pelham, NH, USA). Lipid standards PE 15:0/18:1(+2H7) (1-pentadecanoyl-2-oleoyl(d7)-sn-glycero-3-phosphoethanolamine) and LPC 16:0(+2H3) (1-palmitoyl-d3-sn-glycero-3-phosphocholine) were obtained from Avanti Polar Lipids (Alabaster, AL, USA). A FAME standard mixture, with equal amounts of FAME 16:0, 18:0, 16:1, 18:1, 18:2, 18:3n−3, 18n−6, 20:3, 20:4, 20:5, 22:4, 22:5n−3, 22:5n−6 and 22:6, was obtained from Larodan Fine Chemicals (Solna, Sweden).

Quantitative Lipidomics Analysis Protocol

The solvents n-hexane, 2-propanol (iPrOH), formic acid (FA), acetonitrile (ACN), and methanol (MeOH) of ultrahigh-performance LC/MS grade were purchased from Biosolve (Valkenswaard, Netherlands). Water was produced in-house with a Barnstead GenPure System from Thermo Scientific (Waltham, MA, USA). Methyl tert-butyl ether (MTBE) of pro analysis quality was obtained from Carl Roth (Karlsruhe, Germany). 2-Butanol of extra-pure quality was purchased from Thermo Fisher Scientific (Waltham, MA, USA). 2,6-Di-tert-butyl-4-methylphenol (BHT) of gas chromatography quality and EDTA of ACS reagent grade were purchased from Sigma-Aldrich (St Louis, MO, USA). Ethyl acetate and chloroform (CHCl3) of LC grade, zinc sulfate heptahydrate of pro analysis quality, and phosphate-buffered saline (PBS) of liquid, sterile-filtered, suitable-for-cell-culture grade were obtained from Merck Chemicals (Darmstadt, Germany). Tris (hydroxymethyl)aminomethane–HCl buffer was obtained from Bioanalytic (Umkirch, Germany). Unlabeled and deuterium-labeled PUFA and eicosanoid standards were purchased from Cayman Chemicals (Ann Arbor, MI, USA); the detailed information can be found in Table S1.
