Continuous Microfluidic Synthesis of Lipid Nanoparticles

Ethanol (99.9%), cholesterol (Chol, Sigma Grade ≥ 99%) from sheep’s wool, dipalmitoylphosphatidylcholine (DPPC, >99%), octadecylamine (ODA, ≥99.0%, stearylamine), and polyoxyethylene (40) stearate (PEG-40) were obtained from Sigma Aldrich, Gillingham, UK. PHOSPHOLIPON®90G (purified phosphatidylcholine, or PC, from soybean lecithin) was kindly provided as a gift by Phospholipid GmbH (Lipoid, Ludwigshafen, Germany). Syringe pumps (AL-1010) were purchased from World Precision Instruments (Hertfordshire, UK), and 20 mL BD-Plastipak syringes with luer lock connectors were obtained from Fisher Scientific (Loughborough, UK). Male luer lock rings, polytetrafluoroethylene (PTFE) tubing, and magnetic stirrers (UC152D) were supplied by Cole-Parmer (St. Neots, UK). The tubing that was employed to connect the outlet port of the millireactor to the collection vial was 21.5 cm long (inner diameter: 0.5 mm; outer diameter: 1.6 mm) and was purchased from Cole-Parmer (St. Neots, UK).

Sodium hydroxide (NaOH) was purchased from POCH S.A. (Gliwice, Poland). Methanol was sourced from Merck (Darmstadt, Germany). Mueller-Hinton agar was supplied by Oxoid (Hampshire, UK). All chemicals had an analytical purity and were used without further purification.

Biophysical Characterization of Cytotoxins

Egg yolk L-α-phosphatidylcholine (PC), cardiolipin (CL) from E.coli, bovine brain L-α-phosphatidyl-L-serine (PS), dimiristoylphosphatidylcholine (DMPC), dipalmitoylphosphatidylcholine (DPPC), dimiristoylphosphatidylserine (DMPS), dipalmitoylphosphatidylserine (DPPS), the EPR spin labeled probes 5-doxylstearic acid (5-DSA), and potassium ferricyanide were all purchased from Sigma Chemical Co. (St. Louis, MO). All phospholipids were further purified on silica columns. Naja naja oxiana crude venom was obtained as a gift from Prof. L. Ya. Yukelson (Institute of Biochemistry, Uzbekistan Academy of Sciences). Cytotoxins CTII and CTI were isolated from 500 mg of crude venom according to a previously published procedure [19 (link)]. Cytotoxins were further purified by cation exchange HPLC by using a SCX 83-C-13-ET1 Hydropore column as previously described [20 (link)]. Pyrene, perylene and erythrosine (Sigma-Aldrich, UK) were used as phosphorescent probes while ferrocene (Sigma Chemical Co., St. Louis, MO) was used as a quencher of erythrosine phosphorescence.

Lipid Binding Assay for Pore-Forming Toxin

The analysis was performed essentially as described earlier [33 (link)] with modifications. The following lipids were used: semisynthetic bovine brain sphingomyelin (SM), C16-ceramide, cholesterol, dioleoylphosphatidylcholine (DOPC), dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylethanolamine (DPPE) (all from Sigma). The lipids were dissolved in chloroform:methanol:H2O (1:1:0.3) and 1 μl of the solution containing 25–500 pmoles of lipid was spotted onto a 0.45-μm nitrocellulose membrane. The membrane was pressed with a hot block at 60°C for 5 s according to Taki and Ishikawa [34 (link)], blocked with 1% gelatin and 1% polyvinylpyrrolidone and incubated for 45 min at 20°C with 1 μg/ml GST-PFO in PBS buffer containing 0.03% Tween-20. After washing, the membrane was exposed for 45 min to chicken anti-GST IgY-peroxidase (Rockland). Immunoreactive spots were visualized with the SuperSignal West Pico chemiluminescence substrate (Pierce).
