Quantifying Inflammatory and Coagulation Markers

Serum and soluble inflammatory and coagulation markers were obtained using commercially available kits. Assays were performed according to the manufacture’s protocols. The following markers were measured in blood: pro-inflammatory cytokines serum IL-6 (Human IL-6 Qunatikine HS Elisa Kit), serum IL-18 (Human IL-18/IL-1 F4 ELISA), soluble TNF-α receptor-1 (TNFR1) (Human sTNF RI/TNFRSF1A Quantikine ELISA Kit), and soluble TNF-α receptor-2 (TNFR2) (Human sTNF RII/TNFRSF1B Quantikine ELISA Kit), Interferon gamma-induced protein-10 (IP-10)(Human CXCL10/IP-10 Quantikine ELISA Kit), soluble CD14 (Human CD14 Quantikine ELISA Kit) (all R&D System, Minneapolis, MN, USA) and pro-inflammatory protein C-reactive protein (CRP) (EIA kit, Cayman Chemical Company, Ann Arbor, MI, USA), Serum Amyloid A (SAA) (ELISA, Assaypro, St. Charles, MO, USA) and were also measured. Inflammatory index is a calculated parameter: 0.333*log (IL-6) + 0.666*log (sTNFR1). The following markers of thrombosis and coagulation were also measured: Plasminogen activator inhibitor-1 (PAI-1) (AssayMax Human PAI-1 ELISA kit, Assaypro, St. Charles, MO, USA), D-dimer (ELISA ZYMUTEST DDimer, ANIARA, West Chester, OH, USA) and Fibrinogen (immunoperoxidase assay, GenWay, San Diego, CA, USA).

Cytokine Production in HaCaT Cells

IL-1α, IL-1β and IL-18 productions in HaCaT cells were evaluated by using human IL-1alpha Platinum ELISA (BMS243/2, eBiosciences), human IL-1 β/IL-1F2 Quantikine HS ELISA (HSLB00C, R&D systems), human IL-18/IL-1F4 ELISA (7620, R&D systems) kit, respectively. For dose-dependent assay, cells were plated in 12-well plates at a density of 1×105 cells/well and treated with 0.5, 2 or 10 µM sodium arsenite in DMEM medium for 24 h and the media were collected and analyzed. For time dependent assay, cells were exposed to 2 µM sodium arsenite for 24, 48 or 72 h, respectively. The media were collected for ELISA assay. IL-1α, IL-1β and IL-18 productions in the mouse skin were evaluated by using mouse IL-1alpha Platinum ELISA (BMS611, eBioscience), mouse IL-1 β ELISA Kit (ELM-ILb-CL, Rabiotech), mouse IL-18 platinum ELISA kit (BMS618/3, eBioscience), respectively. Skin tissues were harvested and homogenized in extraction buffer (containing 10 mM Tris PH7.4, 150 mM NaCl, 1% Triton X-100). The homogenates were centrifuged at 10,000×g for 20 min at 4°C and the supernatants were used for the determination of cytokine levels by ELISA assay per the manufacturer's instruction. All the assays were done in triplicate. The adjusted quantities were presented as an average of the data ± standard error of mean (SEM).

Cytokine Measurement in Mouse Samples

Tissue samples were treated as described in SDS-PAGE and Western Blot. Blood collected from mice was transferred in heparin (Ratiopharm GmbH, Ulm, Germany) prewashed micro centrifuge tubes, incubated for 15 minutes at room temperature, and centrifuged for 15 minutes at 2000 Â g.
The following enzyme-linked immunosorbent assay (ELISA) kits were used: mouse Il1b/Il-1F2 DuoSet (DY401), mouse TNF-a DuoSet (DY410), mouse CCL2/JE/MCP-1 DuoSet (DY479), human IL18/IL1F4 ELISA (7620), and human IL1b/IL1F2 DuoSet (DY201), all from R&D Systems (Wiesbaden, Germany), and mouse Il18 ELISA kit (7625; MBL International, Woburn, MA).
