The McMaster Animal Research Ethics Board approved all murine experiments. Five-week-old female NOD.Cg-Rag1tm1MomIl2rgtm1Wjl/SzJ (NRG) mice were purchased from The Jackson Laboratory (Bar Harbor, ME) or bred in-house. Seven to eleven-week-old male NRG mice were injected with 0.5 × 106 NALM6-effLuc cells intravenously. A single doses of engineered T cells was administered after 4 days of tumor growth. Tumor burden was monitored through bioluminescent imaging as we have described previously.11 (link),29 (link) Mice were injected intraperitoneally with D-Luciferin solution (15 mg/ml; Perkin Elmer; Waltham, MA) at a dose of 10uL D-Luciferin solution/gram of body weight 14 min prior to dorsal and ventral imaging using an IVIS Spectrum (Caliper Life Sciences; Waltham, MA). Images were analyzed using Living Image Software v4.2 for MacOSX (Perkin Elmer) and dorsal and ventral radiance was summed. Termination criteria included moribundity or hind limb paralysis. In all cases animal treatment strictly adhered to McMaster Animal Research Ethics Board instructions and guidelines.

AML PDX Engraftment in NRG Mice
