Pesticide Biodegradation Assay Protocol

Technical-grade permethrin, deltamethrin, bifenthrin, fenpropathrin, cyhalothrin, and beta-cypermethrin were obtained from Sigma-Aldrich, USA. Chromatographic grade acetonitrile was purchased from Fisher Scientific, USA. All other reagents used here were obtained from Sigma-Aldrich, USA. The chemicals were dissolved in acetone or dimethyl sulfoxide (DMSO) at a stock concentration of 10 g·L−1, and stored in dark bottles at 4°C prior to use.
The mineral salt medium (MSM) containing 2 g (NH4)2SO4, 0.2 g MgSO4·7H2O, 0.01 g CaCl2·2H2O, 0.001 g FeSO4·7H2O, 1.5 g Na2HPO4·12H2O, and 1.5 g KH2PO4 per liter was used for biodegradation assays in the study. The Luria-Bertani medium (LB) containing 10 g tryptone, 5 g yeast extract and 10 g NaCl per liter was used for incubating Acinetobacter baumannii ZH-14. Both of the media were autoclaved at 121°C for 20 min. The pH of the media was adjusted to 7.0.

Isolation and Characterization of Carboxylesterases

P. synxantha PS1 was isolated from oil well-produced water (Shengli Oilfield, Shandong Province, China). E. coli DH5α (Invitrogen, USA) and plasmid pMD19-T (TaKaRa, Japan) were used for gene cloning and sequencing of AT-containing and AT-truncated carboxylesterases. Plasmid pET-28a (Novagen, USA) was used as a vector to construct the protein expression plasmid in E. coli BL21 (DE3). The substrates fenpropathrin, cypermethrin, fenvalerate, bifenthrin, and p-nitrophenyl esters were purchased from Sigma (St. Louis, MO, USA). All other chemicals were of analytical grade and purchased from local markets.

Technical-grade permethrin, cyhalothrin, β-cypermethrin, deltamethrin, fenpropathrin, and bifenthrin were purchased from Sigma–Aldrich, United States and high-performance liquid chromatography (HPLC)-grade acetonitrile was purchased from Fisher Scientific, United States. All other chemicals and solvents were of analytical grade. SPs were dissolved in dimethyl sulfoxide (DMSO) or acetone at a stock concentration of 10 g⋅L-1, and stored in dark bottles at 4°C.

Pesticide Residue Analysis Protocol

Pesticide reference standards including malathion, diazinon, profenofos, chlorpyrifos, chlorpyrifos-methyl, omethoate, dimethoate, cypermethrin, deltamethrin, fenpropathrin, permethrin, tetramethrin, cyfluthrin, fenvlarate, lamdacyhalothrin , lindane, heptachlor, aldrin, heptachlor epoxide, mixture endosulfan, dieldrin, endrin, o,P-DDT, carbaryl, carbofuran, bendiocarb, atrazine, imidacloprid, thiomethoxam and pendimethalin were purchased from Sigma Aldrich GmbH (Augsburg, Germany), Supelco and Bayer Crop Science , with certified purity ranging from 95% to 99%. Acetonitrile, acetone, sodium chloride, toluene, acetic acid and anhydrous magnesium sulfate were obtained from Scharlau (Barcelona, Spain). QuEChERS Finisterre micro centrifuge tube containing two or more of primary secondary amine (PSA), anhydrous magnesium sulfate (MgSO4) and graphed carbon black (GCB) with details of TR-Q2035 (150mg MgSO4, 25mg PSA, 2.5mg GCB) and TR-Q2015 (150mg MgSO4, 25mg PSA) were obtained from Teknokroma (Barcelona, Spain). Piperonyl Butoxide (PPB) with certified purity 95% used as internal standard was obtained from Sigma Aldrich GmbH.
All the organic solvents used were high performance liquid chromatography (HPLC) grade.

Insecticide Preparation and Dilution Protocol

Tested insecticides were analytical grade and included Bifenthrin (99.8%), Dimethoate (99.8%), fenpropathrin (99.1%), imidacloprid (99.9%), cyantraniliprole (98.0%), sulfoxaflor (99.5%), flupyradifurone (99.5%), and spinetoram (a mixture of 76.2% spinetoram J and 21.0% spintoram L) representing several insecticide classes (Table 1). Dimethoate, fenpropathrin, and imidacloprid were obtained from Sigma-Aldrich (St. Louis, Missouri). Bifenthrin, cyantraniliprole, sulfoxaflor, flupyradifurone, and spinetoram were obtained from Chem Service, Inc. (West Chester, Pennsylvania). Each insecticide was stored according to the manufacturer's recommendation. Serial dilutions of technical insecticide in acetone (ACS grade, 99.5%, Fisher Scientific, Hampton, New Hampshire) were made to generate test concentrations ranging from 0.0001 to 10,000 ng/µL. All insecticide solutions were kept at -20 °C for a maximum of 30 d and new solutions were prepared thereafter.
