Quantification of Cannabinoids in Serum

Reference standards for THC-d3, THC-OH, THC-OH-d3, THC-COOH-d9, CBD, CBD-d3, CBDA, CBN-d3, CBNA, CBG, CBGA, CBC, CBCA, CBL, THCV, THCVA, CBDV and CBDVA were purchased from Cerilliant (Round Rock, TX, USA), CBG-d9 and CBC-d9 from Cayman Chemical Co. (Ann Arbor, MI, USA) via LGC Standards (Wesel, Germany), CBN, THC and THC-COOH from LGC Standards and THCAA from THC Pharm (Frankfurt a. M., Germany). All standards were certified reference material and had a purity of at least 99.0%, except for THC-OH-d3, THCAA, CBC, CBC-d9, CBCA, CBL, THCVA, CBDV and CBDVA (97.7%), THC-d3 (96.7%), and CBG-d9 (95.0%). All chemicals were obtained from Carl Roth (Karlsruhe, Germany), except for methanol (from Fisher Scientific (Loughborough, UK)) and acetonitrile (from AppliChem (Darmstadt, Germany)). Chemicals used in LC-MS/MS measurement processes were LC-MS grade. All other chemicals were HPLC grade, except for acetone and dichloromethane, which had a purity of 99.8% and 99.5%, respectively. Blank human serum used for calibration was obtained from voluntary healthy blood donors via blood donation service and was tested to be drug-free.

Senicapoc Quantification in Biological Samples

Senicapoc was obtained from Sigma-Aldrich (Schnelldorf, Germany). Diazepam-D5 and 11-hydroxy-Δ⁹-tetrahydrocannabinol-D3 (THC-OH-D3) were purchased from Cerilliant (Round Rock, Texas). Methanol (MeOH), acetonitrile (MeCN), dimethyl sulphoxide (DMSO) and formic acid (FA) were obtained from Sigma-Aldrich. Water was purified using a Direct-Q 3 apparatus (Millipore, Bedford, MA).
Stock solutions of Senicapoc (1 mg/mL) were prepared in DMSO. Separate stock solutions (0.1 or 1 mg/mL) of the deuterated substances were obtained in MeOH. Standard solutions to spike the samples and prepare the calibrants were prepared by diluting the stock solutions with MeOH. An IS solution containing 0.05 μg/mL diazepam-D5 and 0.5 μg/mL THC-OH-D3 was prepared in MeOH. Mobile phases A and B consisted of 0.1 % FA and 0.1 % FA in MeCN, respectively.

Quantitative Analysis of Cannabinoids

THC, THC-OH, THC-COOH and CBD at 1 mg/mL, and THCCOOH-glucuronide and the internal standards (IStd) THC-d3, THC-OH-d3 and THCCOOH-d3 at 0.1 mg/mL in methanol were purchased from Cerilliant (Round Rock, TX, USA). THC-diOH and THC-diOH-d6 at 0.1 mg/mL and THC-glucuronide at 0.01 mg/mL in methanol were from ElSohly Laboratories (Oxford, MS, USA). Solid phase extraction (SPE) mixed-mode cation exchange cartridges (Strata Drug X-C, 3 mL, 60 mg) were purchased from Phenomenex (Torrance, CA, USA). Formic acid was obtained from Pharmco-Aaper (Shelbyville, KY, USA). Methanol, acetonitrile, dichloromethane, and isopropanol were acquired from Fisher Scientific (Pittsburgh, PA, USA). Solvents for the extraction were high performance liquid chromatography (HPLC) grade and for the chromatographic instrument were liquid chromatography-mass spectrometry (LC-MS) grade.
