Cell Cycle and Apoptosis Analysis of B-NHL Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
For cell cycle analysis, B-NHL cells were treated with CBL0137 (0.5 μM, 1.0 μM, 1.5 μM, and 2.0 μM) for 24 h and stained with PI/RNase (KeyGEN BioTECH, China) for 30 min. An Annexin V-FITC/PI Apoptosis Kit from Vazyme was used for apoptosis analysis. Flow cytometry was used for detection, and the data were analyzed by FlowJo software.

Blue Light-Induced Apoptosis Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Referring to the previously reported method,42 UMUC‐3 and 5637 cells were inoculated into 6‐well plates at 5.0 × 105 cells per well, respectively, and incubated with packaged lentiviruses paCas9‐Survivin and paProtacL‐Survivin separately or in combination for 48 h, and then followed by blue light irradiation (1.2 W/m2). After 48 h of incubation under blue light irradiation, the apoptosis rate in the above‐treated cells was measured by the flow cytometry (Beckman Coulter) via an Annexin V‐FITC/PI Apoptosis Kit (Vazyme) in relevance to the manufacturer's protocol. Each experiment was repeated three times.

Quantifying Cellular Apoptosis by Annexin V-FITC/PI Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell apoptosis was assessed using an annexin V-FITC/PI apoptosis kit (Vazyme Biotech Co.). A total of 10,000 cells/well were seeded in 6-well plates and treated with germacrone for 48 h. Then, the cells were harvested and washed twice in cold PBS and resuspended in 100 μL binding buffer. The cell suspension was mixed with 5 μL annexin V-FITC and 5 μL PI staining solution for 10 min in the dark at room temperature. The cells were resuspended in 400 μL binding buffer and were analyzed by flow cytometry (Becton Dickinson FACSCanto II, USA) within 1 h.

Annexin V-FITC/PI Apoptosis Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
An annexin V-FITC/PI apoptosis kit (Vazyme, Nanjing, China) was used to analyse apoptosis. Following transfection for 48 h, 5 μl annexin-FITC and 5 μl PI were added to H1299 and A549 cells for 20 min in the dark at room temperature. Apoptotic cells were analysed within one hour via using a flow cytometer (Becton Dickinson, CA, USA).

Quantification of Cell Apoptosis

Check if the same lab product or an alternative is used in the 5 most similar protocols
The AnnexinV-FITC/PI apoptosis kit (Vazyme Biotech Co.) was used to assess cell apoptosis. Cells were first collected and then washed twice in ice-cold PBS, then resuspended in 100 ul binding buffer. We added 5 μl Annexin V-FITC and 5 μl PI staining solution to the suspension for 10 min at room temperature in the dark. We added 400 μl binding buffer to each sample, followed by detection by flow cytometry (Becton Dickinson FACSCanto II, USA) within 1 h.

Apoptosis Assay with Chaetocin

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Annexin V-FITC/PI Apoptosis Kit was purchased from Vazyme Biotech (Nanjing, China). TE-1 and KYSE150 cells were seeded in 6-well plates at a density of 2 × 106 cells/well, and treated with chaetocin at different concentrations (0, 0.2, 0.4, and 0.8 μM) for 24 h in the presence or absence of NAC (5 mM) or z-VAD-fmk (10 μM) before collection. The cells were analyzed by flow cytometry as previously described [25 (link)].

Measuring Apoptosis in Cultured Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
After sucking out the culture medium, cells were rinsed with PBS for three times. Then, the cells were digested with the Trypsin (ThermoFisher, #15050065) without EDTA Solution and centrifuged in 1000 g for 5 min. Apoptotic cells stained with Annexin V-FITC/PI apoptosis kit (Vazyme, #A211-01), and subsequently detected using flow cytometry.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
