N-Glycan Release and Liquid Chromatography

For N-glycan release and analysis by liquid chromatography, the following materials and reagents were used. AcroPrep Advance 96-filter plates and 10 kDa MWCO microcentrifuge filtration tubes were acquired from Pall Life sciences (USA). Polypropylene 2 mL deep 96-well blocks were purchased from Fisher Scientific (Ireland) and 0.45 μm Millex-LH filters from Millipore (Ireland). Plate seals were obtained from Cruinn (Ireland) and silicone sealing mats from Phenomenex (Germany). Protogel was purchased from National Diagnostics (UK) and ammonium hydroxide solution was acquired from Fluka (Ireland). Formic acid and ammonium persulfate (APS) were obtained from VWR chemicals (Ireland). Ultrapure water was filtered through an arium® ProUV system (Sartorius, Germany). HPLC grade methanol, ethanol, acetonitrile, xylene and water were acquired from Fisher Scientific (Ireland). PNGase F was purchased from New England Biolabs. All other reagents were purchased from Sigma (including N,N,N′,N′-tetramethylethane-1,2-diamine (TEMED), iodoacetamide (IAA), dithiothreitol (DTT) and 2-aminobenzamide (2-AB)) unless otherwise specified.

Intrahippocampal Pilocarpine Administration Protocol

The potent and selective GSK-3β inhibitor TCS2002 was purchased from Tocris Bioscience (Bristol, United Kingdom) and the chemoconvulsant pilocarpine.HCl from Sigma (St Louis, MO, USA). All other chemicals were analytical reagent grade and were obtained from Merck (Darmstadt, Germany). Aqueous solutions were prepared freshly, with water purified by an Arium pro UV system (Sartorius Stedim Biotech GmbH, Goettingen, Germany). For the in vivo experiments with intrahippocampal administration of pilocarpine, stock solutions were prepared in ultrapure water, diluted with modified Ringer's solution (147 mM NaCl; 4 mM KCl; 2.3 mM CaCl2), previously filtered through a membrane filter (0.2 µm diameter) and directly delivered into the hippocampus through perfusion of a microdialysis probe. Prior to the systemic administration, TCS2002 was dissolved in propylene glycol:saline (50:50).

Evaluation of Neuroprotective Compounds

Indirubin was obtained from Sigma-Aldrich Chemicals (Bornem, Belgium), BIO-acetoxime was purchased from Tocris Bioscience (Bristol, United Kingdom) and chosen because of its high inhibitory activity towards GSK-3, roscovitine from Selleckchem (Houston, USA), pilocarpine.HCl and PTZ from Sigma (St Louis, MO, USA). All other chemicals were analytical reagent grade and were obtained from Merck (Darmstadt, Germany). Aqueous solutions were prepared in fresh water purified by an arium pro UV system (Sartorius Stedim Biotech GmbH, Goettingen, Germany). Prior to their systemic administration in rodents, Indirubin and BIOacetoxime were dissolved in propyleenglycol:saline (50:50). Since the metabolic and chemical stability of the compounds is unknown, solutions were freshly prepared and syringes (protected from light) were filled ex tempore to minimize the influence of possible degradation.
