Photosynthetic Pigment Quantification

Cell samples were harvested by centrifugation (8000 g, 15 min), and the extraction of photosynthetic pigments was performed with absolute methanol according to [21 (link)]. The extracts were manipulated under dim green light to avoid pigment photo-degradation and were measured with a Pharmacia Ultrospec 2000 UV-Vis spectrophotometer (1-nm bandwidth; Amersham Biosciences, Piscataway, NJ, USA) at 665 nm (Chl a), 653 nm (Chl b), 470 nm (Cars) and 750 nm (background disturbance). Pigment concentrations were quantified as reported in Wellburn [91 (link)] and expressed as μgpigment 10−6 cells.

Quantifying Photosynthetic Pigments in Leaves

Check if the same lab product or an alternative is used in the 5 most similar protocols
Photosynthetic pigments were extracted from pre-weighted small round pieces of leaflets and stipules with a surface area of 28.27 mm 2 . Extraction was performed with 100% methanol for 10 min at 80 °C and pigment concentration was determined according to Wellburn [53] using a Pharmacia Ultrospec 2000 UV-Vis spectrophotometer (1 nm bandwidth) (Amersham Biosciences, Piscataway, NJ, USA). Pigments were quantified both on surface area unit and on fresh weight unit basis. At least four replicates were analysed for both leaflets and stipules.

Photosynthetic Pigment Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
For photosynthetic pigment analysis, cell samples were col-lected by centrifugation after 0, 7, 17, 21, and 25 days of cultivation. Extraction of photosynthetic pigments was per-formed according to Baldisserotto et al. (2014) . The extracts were measured with a Pharmacia Ultrospec 2000 UV-Vis spectrophotometer (1-nm bandwidth; Amersham Biosciences, Piscataway, NJ, USA) at 666 (chlorophyll a (Chla)), 653 (chlorophyll b (Chlb)), and 470 nm (carotenoids (Car)). Quantification was performed according to equations reported in Wellburn (1994) .

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
