Biomarker Measurement in Euthanized Subjects

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected at euthanasia. Serum active GLP-1 concentration was measured using an insulin enzyme-linked immunosorbent assay (ELISA) kit (IBL, Osaka, Japan). In addition, the serum stromal cell-derived factor-1α (SDF-1α) and platelet-derived growth factor (PDGF) concentration was measured using ELISA kits DSA00 and MBB00, respectively (R&D Systems, Minneapolis, MN, USA). Urinary 8-hydroxy-2’-deoxyguanosine (8-OHdG) concentration was measured using a kit (JaICA, Shizuoka, Japan). Urinary creatinine levels were also measured using a kit (Wako, Osaka, Japan) to adjust the urine concentration, as previously described [23 (link)].

Quantification of TGF-β1 and PDGF-BB

Check if the same lab product or an alternative is used in the 5 most similar protocols
We determined the concentration of active TGF-β1 in the serum using the TGF-β1 ELISA Development kit (human: R&D Systems, DB100B; mouse: R&D Systems, MB100B) and PDGF-BB using PDGF-BB ELISA Development kit (human: R&D Systems, DBB00; mouse: R&D Systems, MBB00) following the manufacturer’s instructions.

Quantitative Tissue Biomarker Assay

Harvested lung and kidney tissues were snap-frozen in liquid nitrogen
and homogenized in RIPA lysis buffer (27 (link)). Blood samples obtained via terminal cardiac punctures were
collected in heparin-coated plasma-separating tubes (BD Microtainer #365985) and
centrifuged for 10 minutes at 1,000 RCF (g) to isolate plasma.
Quantikine ELISA kits were used to measure murine Ang2 (R&D #MANG20),
VEGF (#MMV00), PDGF-BB (#MBB00), and erythropoietin (EPO; #MEP00) levels in
plasma or tissue homogenates.

Quantifying Growth Factors in Tumor Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Enzyme-linked immunosorbent assay was used for measuring human FGF-2, mouse VEGF, and mouse PDGFB protein concentrations. To detect protein levels in tissues and cells, tumor tissues and cells in culture were homogenized in a lysis buffer (C3228, Sigma), containing proteinase inhibitor cocktails (1:100, P8340, Sigma), followed by 15 min centrifugation. The supernatants were transferred to a new collection tube and either immediately used or stored at −20 °C or −80 °C until further analysis. Conditioned media were collected from confluent E0771 and T241 cells at 24 and 48 h. Cell debris was removed by centrifugation and supernatants were stored at −20 °C or −80 °C until analysis. The human FGF-2 (DFB50, R&D systems), mouse VEGF (MMV00), and mouse PDGF-BB ELISA assays (MBB00, R&D Systems) were performed according to the manufacturer’s protocol with the standard curve. Absorbance values were detected at 450 nm using a microplate reader and values were calculated using the formula obtained from the trendline.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
