Cell Viability Assessment via CCK-8 Assay

Cell viability was analyzed using the Cell Counting Kit-8 (CCK-8; Servicebio) in accordance with the CCK-8 assay protocols. Cells were seeded into 96-well plates at a density of 2 × 103 cells/well. When cultured to 0, 24, 48, 72, and 96 h, CCK-8 (10 µL/well) was added to the cells and incubated at 37 °C for 1 h. We measured the absorbance of each well at 450 nm using a microplate reader (Bio-Rad Laboratories, Inc.).

Fluorescent Labeling and Cell Viability Assay

DOX and Cyanine 5.5 (Cy5.5, C41H44N2O14S4) were purchased from Yuanye (Shanghai, China). Caspase 9 antibody, BAX antibody, Ki67 antibody, CD3 antibody, CD4 antibody, and β actin antibody were purchased from Proteintech Company (Wuhan, China). Gp100 was purchased from Affinity Biosciences (Santa Clara, CA, USA). B16 cell lines were donated by the National Engineering Laboratory for AIDS Vaccines at Jilin University (24 (link)), and the use of these cell lines received approval from the institutional review board for research ethics at the China-Japan Union Hospital of Jilin University.

Cell viability was measured using Cell Counting Kit-8 (CCK-8) (Servicebio). In brief, cells were plated in 96-well plates at the density of 1 × 104 cells per well. When cells adhered, different concentrations of gefitinib, erlotinib, or icotinib (Energy Chemical, Shanghai, China) were added into the wells. The same volume of DMSO was added into the control well. After 24 h incubation, 10 μl of CCK-8 reagent was added to each well, and the plates were incubated for an additional 30 min. The absorbance was read at 450 nm. The IC50 value was calculated based on the non-linear regression fit method by GraphPad Prism 9.5.1 software (GraphPad Software, San Diego, CA, USA).

Evaluating IPA Cytotoxicity on Chondrocytes

The cytotoxicity of IPA on chondrocyte viability was quantitated by Cell Counting Kit-8 (CCK-8, Servicebio, G4103). Chondrocytes were seeded in 96-well plates at 5 × 103 cells per well. Then IPA was added at concentrations of 0, 10, 20, 40, 80, 160, 320 and 640 µM. 10 µL of CCK-8 solution was added to each well at 0, 24, 48, 72 and 96 h, and incubated at 37 °C for 2 h. The absorbance at a wavelength of 450 nm was determined using a microplate reader (EnVision, PerkinElmer, USA).
