Secondary antibodies used (1 : 2000 for immunofluorescence) were conjugated with Alexa 488, Alexa 568 or Alexa 647 (Invitrogen) and had minimal cross-reactivity to other species.

Andro (365645) and ammonium pyrrolidinedithiocarbamate (APDC; NF-κB inhibitor, P8765) were obtained from Sigma-Aldrich (St. Louis, MO, United States) and were dissolved in dimethyl sulfoxide (DMSO). Cell Counting Kit-8 (CCK-8) was purchased from Beyotime (Shanghai, China). Rabbit anti-Ki67 (ab15580) was purchased from Abcam (Cambridge, MA, United States), rabbit anti-GAPDH (2118) was purchased from Cell Signaling Technology, Inc. (Danvers, MA, United States), rabbit anti-PDCD4 (BM5208) and rabbit anti-EGN (endoglin, BA2227) were both from Boster (Wuhan, China), and rabbit anti-NF-κB p65 (abs131170) and rabbit anti-phospho-NF-κB p65 (Ser536, abs130624) were purchased from Absin (Shanghai, China). All the miRNA mimics and inhibitors and the negative control (NC) RNAs were purchased from RiboBio Co., Ltd. (Guangzhou, China). All the primers were synthesized by Sangon Biotech Co., Ltd. (Shanghai, China).

The following antibodies were obtained from Cell Signaling Technology (Beverly, MA): rabbit anti-CDK2 (2546), rabbit anti-pCDK2-T160 (2561), rabbit anti-pp100-S866/870 (4810), rabbit anti-pIĸKα/β (2078), rabbit anti-IĸKα (2682), rabbit anti-NF-ĸB2 p100/p52 (4882), rabbit anti-NIK (4994), rabbit anti-β-TrCP (D13F10) (4394), and rabbit anti-PCNA (13110). β-TrCP siRNA (sc-37179), p52 siRNA (m) (sc-36043), CDK2 shRNA (m) lentiviral particles (sc-29260-V), NFκB p52 shRNA (m) lentiviral particles (sc-36043-V), goat anti-pNIK-T559 (sc-12957), rabbit anti-Epo (sc-7956), mouse anti-GAPDH (sc-32233), mouse anti-β-actin (sc-47778), were purchased from Santa Cruz Biotechnology (Santa Cruz, CA). Rabbit anti-Epo (ab65394) and rabbit anti-Ki-67 (ab15580) were purchased from Abcam (Cambridge, MA). Other chemicals and organic solvents of the highest available grade were obtained from Sigma-Aldrich. AMPKα1-KO and AMPKα2-KO mice were described elsewhere [55 (link), 56 (link)]. Mice were handled in accordance with study protocols approved by the Institutional Animal Care and Use Committee of University of Oklahoma Health Sciences Center (Oklahoma City, OK).

Rabbit anti-SQSTM1/P62 (D5E2), rabbit anti-LC3A/B (D3U4C), rabbit anti-mTOR (7C10), rabbit anti-p-mTOR (Ser2448), rabbit anti-p-SAPK/JNK (Thr183/Thr185), rabbit anti-SAPK/JNK (56G8), rabbit anti-cyclinD1 (96G2), rabbit anti-p-ERK (Thr202/Thr204), rabbit anti-ERK (137F5), rabbit anti-p-P38 (Thr180/Tyr12), rabbit anti-P38 (D13E1), rabbit anti-p-AKT (Ser473, D9E), and rabbit anti-AKT (C67E7) were from Cell Signaling (Danvers, MA, USA). Rabbit anti-GRP78 (ab21685), rabbit anti-IRE1 (ab37073), rabbit anti-NCX1 (ab151608), and rabbit anti-Ki67 (ab15580) were from Abcam (Cambridge, MA, USA). Mouse anti-GAPDH was from Beyotime Biotechnology (Nanjing, China). NCX1 inhibitor KB-R7943 (Sigma, St Louis, MO, USA, K4144-25MG) was dissolved in DMSO at 10 mM. Autophagy inhibitor 3-methyladenine (Sigma, M9281-100MG) was dissolved in medical injectable water at 50 mM. Chloroquine (Sigma, C6628-25G) was dissolved in medical injectable water at 10 mM. Wortmannin (Beyotime Biotechnology, Nanjing, China) and SP600125 (Sigma, S5567-10MG) were dissolved in DMSO at 10 mM.

Mice at different time-points of reperfusion were euthanized by perfusion with ice-cold saline, and the brains rapidly dissected and stored at −80 °C. As we previously described [5 (link)], TTC staining was performed to demarcate the cerebral infarct and ischemic penumbra. Immunofluorescent (IF) studies were performed as previously described [30 (link)] on 10-μm-thick frozen coronal sections. The following monoclonal antibodies from BD Pharmingen (La Jolla, CA, USA) were used in this study: FITC-conjugated rat anti-mouse CD31 (PECAM-1) (clone MEC13.3; 1:100) and PE-conjugated rat anti-mouse α5(CD49e) (clone 5H10-27; 1:100). The rabbit anti-fibronectin (F3648) was purchased from Sigma (St. Louis, MO, USA; 1:400). The rabbit anti-Ki67 (ab15580) was obtained from Abcam (Cambridge, MA, USA; 1:800). The rabbit anti-CD31 (PECAM-1) (250590) antibody was obtained from Abbiotec (San Diego, CA, USA; 1:200). The rabbit anti-integrin β3 (MBS8508681) antibody was purchased from MyBioSource (San Diego, CA, USA; 1:200). The Alexa Fluor 488-conjugated goat anti-rat and anti-rabbit, and Cy-3-conjugated goat anti-rat and anti-rabbit secondary antibodies were obtained from EarthOx (Millbrae, CA, USA).
