The following analytical grade or chemically pure reagents were purchased from commercial sources for this study: 30% hydrogen peroxide (H2O2), sodium acetate trihydrate (CH3CO2Na·3H2O), ferrous sulfate heptahydrate (FeSO4·7H2O), sodium nitrite (NaNO2), disodium hydrogen phosphate dodecahydrate (Na2HPO4·12H2O), sodium dihydrogen phosphate dihydrate (NaH2PO4·2H2O), ferric chloride hexahydrate (FeCl3·6H2O), anhydrous sodium carbonate (Na2CO3), potassium acetate (CH3CO2K), aluminum chloride (AlCl3). Potassium ferricyanide (K3[Fe(CN)6]), salicylic acid sodium, gallic acid monohydrate, Folin-ciocalteu reagent, 2,6-di-tert-butylphenol, Rutin (C27H30O16), t-butyl terephthalate phenol, Tris buffer, 2,4,6-Tri(2-pyridyl)-1,3,5-triazine (TPTA) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) were obtained from Sigma chemicals.

Antioxidant Evaluation of Vaccinium myrtillus Leaves

Vaccinium myrtillus dry leaves were purchased from Bidah Chaumel (Murcia, Spain). They proceed from an organic field located in Huelva (Spain), and were collected in June 2022. Fresh leaves were air dried in dark conditions at room temperature in order to reach a moisture of 10.5%. They were grounded to an average particle size of 0.8 mm and stored at −20 °C until the extraction.

A gram stain kit was obtained from YD Diagnostics (Korea). An API 50 CHL kit was obtained from Bio Merieux Ltd. (France). 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid was obtained from Fluka (Germany). Lactic acid was obtained from Konto (Japan). BCP agar was obtained from Aiken (Japan). Skim milk was obtained from Seoul Dairy Co-op. (Korea). Chloroform was obtained from JT Baker (USA). NaOH pellets were obtained from Mallinckrodt (USA). Finally, the sterilized 0.45 μm filters were obtained from Millipore (Ireland).

Antioxidant Assays and Phenolic Compound Analysis

Ultrapure water was prepared by the Milli-Q system (Millipore, Bedford, OH, USA). All solutions prepared for HPLC were passed through 0.45 μm nylon filters before use. All other chemicals were of analytical reagent grade.

Antimicrobial Activity Evaluation Protocol

Dimethyl sulfoxide (DMSO) of 99.90% purity was obtained from Sigma-Aldrich, Germany; while Amoxicillin and Penicillin were purchased from Thermo Fisher Scientific, USA. All the chemicals were of analytical grade unless otherwise stated, and used without further purification. Deionized water was used for preparing the reagents and solutions, carrying out extractions and chromatographic analyses.
Standard strains of Staphylococcus aureus (CCM 4223) and Escherichia coli (CCM 3954) were obtained from the Czech collection of microorganisms (Brno, Czech Republic). The strains were cultivated over night at 37 °C on 5% Columbia blood agar (Sigma-Aldrich, Germany).

Ferric chloride. 6H2O (lot 9n005099n) was obtained from Carlo Erba (Barcelona, Spain). 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) (lot p31b064) was obtained from Alfa Aesar (Haverhill, MA, USA). Annatto seeds were purchased from a farmers market of Medellín city (Colombia), which were sun-dried until reaching a moisture content of 10.58 ± 0.98%.
