PDGFB Knockdown in Mesenchymal Stem Cells

Validated siRNA duplex constructs targeted against human PDGFB were purchased from Sigma Aldrich (SASI_Hs01_00121961, SASI_Hs01_00121962, (St. Louis, MO, USA). Mission siRNA Transfection reagent (Sigma Aldrich) was used to reverse transfect 10 nM of siRNA to MSC, in a 24-well format following manufacturer’s protocols. A universal negative siRNA non-targeting control (Sigma Aldrich) was utilized to rule out effects from transfection protocols.
