Mitochondrial Membrane Potential Assay

MMP is key toward maintaining mitochondrial function, which was measured with a commercial kit of JC-1 (Beyotime). The procedure was performed according to the manufacturer’s instructions. Mitochondria from liver tissue were separated using a tissue mitochondria extraction kit (Beyotime) and were then incubated with JC-1 solution for 30 min at 37°C. The fluorescence in different groups was measured with a micro-plate reader (Bio-Rad, Hercules, CA, USA). The MMP percentage was finally calculated in order to evaluate the loss of MMP in the liver.

Mitochondrial Membrane Potential Measurement

Check if the same lab product or an alternative is used in the 5 most similar protocols
The mitochondrial membrane potential (ΔΨm) was determined using a JC-1 probe kit (Beyotime, Shanghai, China). Firstly, a small section (50 mg) of liver was excised and weighted immediately after rat sacrifice. Liver mitochondria was obtained by a tissue mitochondria extraction kit (Beyotime, Shanghai, China). Thereafter, the purified mitochondria were stained with a 1× JC-1 working solution, and samples were detected by a fluorescence microplate. ΔΨm was calculated as the ratio of red fluorescence (JC-1 aggregates) to green fluorescence (JC-1 monomers).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
