Lippia sidoides EO was purchased from PRONAT (Produtos Naturais do Nordeste LTDA, Horizonte, Ceará, Brazil). The constituents of the NLCs were Gelucire 50/13 (Gattefossé, Saint-Priest, France), oleic acid (LabSynth, Diadema, São Paulo, Brazil), Labrasol (Gattefossé, Saint-Priest, France), Span® 80 (Sigma-Aldrich, Darmstadt, Germany), Tween® 80 (LabSynth, Diadema, São Paulo, Brazil), and Poloxamer 188 (Kolliphor P 188 Micro, kindly donated by BASF, Ludwigshafen, Germany). Mixtures of whey protein (Arla Foods Ingredients S.A., Buenos Aires, Argentina), arabic gum (Nexira, São Paulo, Brazil), and colloidal silicon dioxide (Aerosil 200, Evonik Degussa, Essen, Germany) were used as spray-drying carriers. ultrapure water (Direct Q System, Millipore, Merck KGaA, Darmstadt, Germany) was used in the preparation of NLCs. Other chemicals and solvents used were of pharmaceutical or of high performance liquid chromatography (HPLC) grade.

Analgesic and Anti-inflammatory Compound Preparation

The following materials were used: acetic acid and absolute ethyl alcohol (Neon, São Paulo, Brazil), L-glutamic acid (glutamate) (Vetec, Rio de Janeiro, Brazil), capsaicin, cinnamaldehyde, indomethacin (Sigma-Aldrich, St. Louis, MO, USA), ketamine hydrochloride, xylazine hydrochloride (Syntec, São Paulo, Brazil), naloxone hydrochloride (Cristália, São Paulo, Brazil), formalin (Dinâmica, São Paulo, Brazil), sodium hydroxide (Nuclear, São Paulo, Brazil), menthol (A Essência, Santa Catarina, Brazil), and tween 80 (Labsynth, São Paulo, Brazil). The drugs were dissolved in 0.9% NaCl solution (saline) before administration, except cinnamaldehyde (1% tween 80 in saline), glutamate (saline solution at pH 7), and menthol (1.6% absolute ethyl alcohol + 0.01% tween 80 in saline). capsaicin was prepared from a 0.5% capsaicin stock solution dissolved in absolute ethyl alcohol, and the 0.1% solution was prepared at the time of use by mixing the stock solution with tween 80 and 2:1:7 saline solution.

Standardizing Conidial Suspensions of Aspergillus

Conidial suspensions of each strain were individually prepared and had their concentration standardized at 106 conidia/mL according to Wigmann, et al.58 (link). A. carbonarius and A. niger strains were inoculated in MEA- Malt Extract Agar medium (Acumedia) (Malt Extract: 20.0 g, Peptone: 1.0 g, Glucose: 30.0 g, Agar: 20 g, Distilled Water: 1 L) and incubated at 25 °C for 7 days. Conidia were collected by scraping the mycelium from each plate with sterile distilled water and 0.1% Tween 80 (Labsynth). Subsequently, they were filtered and then centrifuged at 11962.6 × g three consecutive times for 15 minutes at 5 °C (Sorvall Legend XTR, Thermo Scientific, Hampton, USA). The final concentration of conidia in each fungal suspension was determined in Neubauer chamber (Sigma-Aldrich).

Mycotoxin-Producing Aspergillus Strain Preparation

Aspergillus parasiticus NRRL 2999 (a strain with the ability to produce mycotoxins such as AFB1, AFB2, AFG1, and AFG2) was kindly donated by the United States Department of Agriculture. This strain was isolated from soil in Uganda as reported by Singh and Cotty (2019) (link). The strain was prepared according to Wigmann et al. (2016) (link) by inoculating it in Malt Extract Agar (MEA; Acumedia, Lansing, Michigan, United States; Malt Extract: 20.0 g; Peptone: 1.0 g; Glucose: 30.0 g; Agar: 20 g; and Distilled Water: 1 L) and incubating it aerobically at 25°C in a BOD incubator for 5 days, at static condition in the dark. Conidia were collected by scraping the mycelium from each plate with sterile distilled water containing 0.1% Tween 80 (Labsynth, Diadema, Brazil). Subsequently, they were filtered and then centrifuged at 11,000 g three consecutive times for 15 min at 4°C (Sorvall Legend XTR, Thermo Scientific, Hampton, Unites States). The final concentration of conidia in the fungal suspension was determined using a Neubauer chamber (Sigma-Aldrich, Darmstadt, Germany). The conidial suspension was diluted and used to reach a final concentration of 5.0 log10 cfu ml−1 in the growth trials (checked by plate count).

Antimicrobial Compounds from Sigma-Aldrich

Both bacteria were gently given by the Laboratory of Applied Biochemistry and Molecular Biology (LBBMM) from the Department of Genetics and Evolution of Federal University of São Carlos (UFSCar), São Paulo, Brazil. Tween 80 (ethoxylated sorbitan monooleate) was obtained from Labsynth Brasil Ltda.
