Phenolic Substrate Extraction and Characterization

Phenolic substrates including ferulic acid, gallic acid, syringaldehyde (SGA) and catechol were purchased from Tokyo Chemical Industry Co., Ltd. 2,2′-Azino-bis(3-ethylbenzthiazoline-6-sulfonate) (ABTS) and Coomassie Brilliant Blue G-250 were obtained from Sigma-Aldrich. DEAE cellulose and Sephacryl (S-100) were acquired from Bio-Rad and Pharmacia, respectively. Basic chemicals, namely malt extract, ferrous sulphate, acrylamide, bis-acrylamide, ammonium persulphate, TEMED (N,N,N′,N′-tetramethyl ethylenediamine), sodium dodecyl sulphate (SDS), Tris buffer, ammonium sulphate, glutamic acid, monopotassium phosphate, copper and zinc sulphate, were obtained from Himedia. The centrifugal concentrators were obtained from Amicon Millipore, while the dialysis tubing was from Thermo Fischer Scientific. The bundle of human hair was picked up from a local salon. The collected hairs belonged to the same person. All other chemicals used were of analytical grade.

Isolation and Purification of C-PC from Cyanobacteria

C-PC was isolated and purified from Plectonema sp. of cyanobacteria as described in our previous reports (13) (link) and BSA was obtained from Hi-media, sodium carbonate, sodium dihydrogen phosphate, sodium bicarbonate, disodium hydrogen phosphate, methylglyoxal, sodium chloride, oxalic acid, nitroblue tetrazolium (NBT), thiobarbituric acid (TBA), trichloroacetic acid (TCA), sodium hydroxide, phosphotungstic acid, sodium dodecyl sulphate (SDS) and sodium chloride were purchased from Hi-Media. Sodium azide (Na 3 N) and ethanol were obtained from Merck and 2,4,6 trinitrobenzene sulphonic acid (TNBS) was purchased from G-Biosciences. Thioflavin-T (ThT), congo red, phenanthrenequinone, 1-anilinonapthalene-8-sulfonic acid, 5,5-dithiobis-2-nitrobenzoic acid (DTNB), 2,2-diphenyl-1-picrylhydrazyl (DPPH), potassium ferricyanide [K 4 Fe(CN) 6 ], ferric chloride (FeCl 3 ), sodium nitroprusside, sulfanilic acid, naphthalene diamine chloride and triphenylphosphine were purchased from Sigma Aldrich. Ethylacetate was purchased from Emplura, and dinitrophenylhydrazine (DNPH) was purchased from Rankem. Guanidium hydrochloride was purchased from SD Fine Chemical Limited. Hydrochloric acid (HCl) was purchased from Fisher Scientific. All other chemicals used in this study were of analytical grade.

Lipophilic Eugenol Solubilization and Quantification

Eugenol, thiobarbituric acid (TBA) and Evans blue were procured from Sigma Aldrich. Hank’s balanced salt solution (HBSS) was procured by Thermo Fisher Scientific. TEP (1, 1, 3, 3–tetra ethoxy propane), sodium dodecyl sulphate (SDS), glacial acetic acid, n-butanol, pyridine, phosphate buffer, hydrogen peroxide (H2O2) and all the other reagents and solvents were purchased from HiMedia Laboratories Pvt. Ltd., Mumbai. Eugenol is lipophilic and therefore it is insoluble in water. Hence it was suspended in 40% propylene glycol (PG) in phosphate buffer (pH 6.8).

Preparation and Characterization of MPTP

MPTP was procured by Sterling biologicals, Ahmedabad while Hank's balanced salt solution (HBSS) was obtained from Invitrogen (Thermo Fisher Scientific), Bangalore. Eugenol, 5,5-dithio-bis-(2nitrobenzoic acid) (DTNB) (Ellman's Reagent) and thiobarbituric acid (TBA) were purchased from Sigma Aldrich (USA). Sodium dodecyl sulphate (SDS), 1,1,3,3-tetraethoxypropane (TEP), trichloroacetic acid, pyridine, n-butanol, glacial acetic acid and all additional solvents and reagents were acquired by HiMedia Laboratories Pvt. Ltd., Mumbai. Eugenol was suspended in 40% propylene glycol in phosphate buffer with pH 6.8 as it is hydrophobic. MPTP is water-soluble so it was dissolved in sterile saline.
