Experiments were conducted as sex unbiased, with randomly assigned 2-5 mice per experimental group. All mice were bred and maintained in a specific-pathogen-free barrier facility with free access to food and water. Both male and female mice were used between 16-36 weeks of age for most experiments, unless specified otherwise. All mouse studies and protocols were approved by the Institutional Animal Care and Use Committee (IACUC) at Albert Einstein College of Medicine. Wild-type C57BL6/J, Rosa26STOP-tdTomato (AI14) (Madisen et al., 2010 (link)), Rosa26-CreERT2 (Ventura et al., 2007 (link)), CD4-Cre (Lee et al., 2001 (link)), Cxcl12DsRed (Ding and Morrison, 2013 (link)), PA-GFP (Victora et al., 2010 (link)), APRIL−/− (Xiao et al., 2008 (link)), Blimp1-YFP (Fooksman et al., 2010 (link)) and B1-8hi (Shih et al., 2002 (link)) mice were used for most experiments and obtained from The Jackson Laboratory. Wild-type C57BL6 mice were used for PC-intrinsic-age-related CXCR4 level studies and obtained from the Charles River Laboratories.

Genetically Modified Mouse Models for Vascular Research

Animal experiments were approved by the Institutional Animal Care and Use Committee of Keio University (protocol no. A2021-003) and were performed in accordance with the Guidelines of Keio University for Animal and Recombinant DNA Experiments (protocol no. D2005-044). Cdh5-BAC-CreERT2 (ref. 39 (link)), Osx-CreERT2 (ref. 8 (link)), Vegfr2-flox40 (link), Dll4-flox41 (link), Col1a1-flox42 (link), CAG-LSL-EGFP43 (link), Vegfr1-BAC-DsRed44 (link), Vegfr2-BAC-GFP44 (link) and Rbpj-flox45 (link) mice had been developed previously. All mice were crossed with C57BL/6J mice more than 8× and maintained. For analyses of phenotypes, mice of both sexes were used. Csf1op/op (stock no. 000231) and Cxcl12+/DsRed (stock no. 0225458) mice were obtained from the Jackson Laboratory. For neonatal experiments of tamoxifen-inducible expression of Cre, 40 μg of 4-hydroxytamoxifen (4OHT, Sigma-Aldrich, catalogue no. H6278) was subcutaneously injected at the indicated timepoints. For adult experiments of tamoxifen-inducible expression of Cre, 1 mg of tamoxifen (Sigma-Aldrich, catalogue no. H5648) was subcutaneously injected at the indicated timepoints.

Standardized Mouse Husbandry and Genetic Strains

All mouse husbandry and experiments were performed in accordance with Stanford Institutional Animal Care and Use Committee (IACUC) guidelines. All mice were housed in a pathogen-free facility. A maximum of 5 adult mice were housed per cage. Animals were monitored daily for normal husbandry and any health concerns. Cages were kept under a 12-hour light and 12-hour dark cycle. Genetic Manipulation of Murine Vascular Biology

For neonatal studies, all Tamoxifen injections to activate Cre activity (6mgs in corn oil) were given intraperitoneally to nursing mothers. Due to mouse genetic inheritance, analyses were compiled from age-matched males and females from multiple litters.
