Riluzole (Selleckchem, Houston, TX, USA) was resuspended in DMSO at 30mg.ml⁻¹ as a 500-fold concentrated stock solution and further diluted in drinking water. Water was replaced every three days. Ceftriaxone (Cayman Chemical, Ann Arbor, MI, USA) was resuspended in PBS at 100 mg.ml⁻¹ and a 100 mg.kg⁻¹ dose was administered daily by intraperitoneal injection. Dosing of investigational agents was initiated after weaning of the animals on day of life ~21. Six mice per group were sacrificed at 7 weeks of age for pathology and RNA extraction, the remaining animals were treated until they reached the humane disease end point described above. Mutant, control, treated and untreated animals were enrolled concurrently, and efforts were made to balance sex distribution.
