Apoptosis Analysis of Tumor Cells

After exposed to H2 for 12 h, tumor cells were transferred and seeded in the cell culture slide. After fixation and washing, cells were blocked for 1 h, then stained with cleaved caspase-3 antibody or Cytochrome C Antibody (Abcam, UK) overnight at 4 °C on a shaker. The cells were counterstained with fluorescein isothiocyanate (FITC, ABCAM, USA) or allophycocyanin (APC, ABCAM, USA) conjugated secondary antibody for 1 h. Finally, the cells were washed with an Immunol staining wash buffer three times before observation by CLSM. Nuclei were stained with Hoechst.

Cytochrome C Release Measurement

Cytosolic Cytochrome C release from the mitochondria was measured by immunoblot of cytoplasmic and mitochondrial lysates. Briefly, whole mitochondria were isolated as above and lysed in 1X CST buffer from HA magnetic beads. Cytoplasmic proteins were isolated by collecting supernatant after incubation of cellular homogenates with HA magnetic beads. The cytoplasmic fraction was cleared with 25 μl additional HA magnetic beads for 3.5 minutes with rotation at 4°C to further purify the cytoplasmic fraction. Lysates were prepared by adding 10X CST containing protease inhibitor to this fraction. Mitochondrial and cytoplasmic fractions were subjected to SDS-PAGE followed by immunoblot with Cytochrome C antibody (Abcam) at 1:500 dilution.
