N-Glycan Purification and Analysis

N-Linked glycans were released from tryptic digests using PNGaseF. Released glycans were separated from deglycosylated peptides using C18 reversed-phase spin columns (Thermo Pierce, Rockford IL). Glycans were desalted using PD MiniTrap G-10 columns (GE Healthcare, Piscataway NJ), as per the manufacturer’s instructions. Glycan samples were analyzed, in triplicate, using HILIC-MS using an Agilent™ 6520 LC-MS system with a chip-cube nanoESI source, as described previously [29 (link)].
