CAR/TCR T-cell Stability Evaluation

Quantifying BMEC Apoptosis via Annexin V-FITC/PI

First, BMEC apoptosis was detected using an Annexin V-FITC and propidium iodide (PI) Apoptosis Detection Kit (Thermo Fisher Scientific). After transfection, 500 μL of binding buffer was used to resuspend the BMECs. Then, 5 μL PI and 5 μL Annexin V-FITC were added to the suspended cells, followed by a 20-minute darkroom incubation period. The samples were washed on BD FACS™ Lyse Wash Assistant (BD FACSCanto™ II and BD FACSCanto™ 10-Color; BD Biosciences). Cytometry was used to assess the degree of cell apoptosis.
