Synthesis and Characterization of Chitosan Nanomaterials

Chitosan (CS) (MW = 70 kDa, degree of deacetylation of 90%) was purchased from Vietnamese Chitosan Co., Ltd. Methacrylic acid (MAA), potassium persulfate (K2S2O8), and acetic acid (CH3COOH) were purchased from Merck, Germany. Glycerol and CaCl2 were purchased from Xilong, China and BHI medium was purchased from Sigma–Aldrich, Saint Louis, MO, USA.
The pectin (P) was extracted from Tiliacora triandra using a heating method [15 ]. The degree of esterification of pectin was 48.36 and its molecular weight was 111.1 kDa.
Nanochitosan (NCS) were prepared by polymerizing Methacrylic acid (MAA) in chitosan (CS) solution according to the method described by Nguyet and de Moura [5 (link),16 ,17 ]. Firstly, chitosan was dissolved in a 0.5% MMA solution for 12 h. The chitosan concentration used in the synthesis was 0.8 w/w%. K2S2O8, with a concentration of 0.6 mmol was then added to the chitosan—methacrylic solution at 70°C for 1 h under stirring. After that, the mixture was cooled at 4 °C. The suspension was centrifuged at 4000 r/min for 30 min and a suspension was obtained [5 (link)]. Chitosan nanoparticles were twice washed in distilled water, producing a chitosan nanoparticles suspension (NCS).

Quantification of Polyphenols and Sugars

All the standard polyphenols, standard simple sugars (glucose, mannose, galactose, cellobiose, and fructose), and standard for vitamin C quantification (ascorbic acid, dehydroascorbic acid, cetrimide, potassium dihydrogen phosphates (KH2PO4), 1,2-phenylenediamine dihydrochloride (OPDA), and orthophthaldehyde (OPA)) were purchased from Sigma–Aldrich (St. Louis MO, USA). The 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), 2,2′-azo-bis-(2-metilpropionamidina) dihidrocloro (AAPH), 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox), Folin–Ciocalteu reagent, fluorescein, and sulfuric acid were purchased from Sigma-Aldrich (St. Louis, MO, USA). The potassium persulfate (K2S2O8) was obtained from Merck (Kenilworth, NJ, USA).

Determination of Antioxidant Compounds

Folin–Ciocalteu’s reagent, potassium persulfate (K2S2O8) and standards of gallic acid were purchased from Merck (Darmstadt, Germany). Anhydrous sodium carbonate (Na2CO3) was acquired from Panreac (Castellar del Vallès, Barcelona, Spain). ABTS diammonium salt (2,2-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid)), acetic acid and standards of Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) were obtained from Sigma-Aldrich (Madrid, Spain). Glycerol was acquired from Labbox (Barcelona, Spain), ultrapure water was obtained using a Milli-Q water system (Millipore, Milford, MA, USA) and ethanol was purchased from Romil Ltd. (Waterbeach, UK). High performance liquid chromatography (HPLC) grade acetonitrile was acquired from Honeywell Research Chemicals (Seelze, Germany), whereas the HPLC standards of hydroxytyrosol, protocatechuic acid, verbascoside, luteolin-7-O-glucoside, apigenin-7-O-glucoside, luteolin, oleuropein and apigenin were purchased from Sigma-Aldrich (Madrid, Spain).

Hemin was purchased from Acros Organics (Geel, Belgium). Ethanol (99.9%) was purchased from Chemethyl A/S (Køge, Denmark), and sodium hydroxide was purchased from Sigma-Aldrich (Renningen, Germany).

Carbohydrate Standards for Oligosaccharide Analysis

The carbohydrates standards for identification and quantification of oligosaccharides were d-xylose, xylobiose, xylotriose, xylotrehalose, and xylopentaose were acquired at Megazymes (Bray, Ireland), while polysaccharides standards for molecular weight were bought from ShodexTM (Munich, Germany) and d-galactose and d-cellobiose from Sigma-Aldrich (St. Louis, MA, USA).
All the consumables and enzymes used for the simulation of the gastrointestinal tract used in this work were purchased from Sigma-Aldrich (St. Louis, MA, USA): α-amylase, bile salts, pork pancreatin, pork pepsin, as well as hydrochloric acid (HCl), sodium hydrogen carbonate (NaHCO3), and sulphuric acid. From Merck (Kenilworth, NJ, USA), potassium persulfate (K2S2O8) was obtained.
For the assessment of antioxidant activities and total polyphenolic compounds, the following reagents were used: Folin-Ciocalteu reagent, fluorescein, ABTS, AAPH, and Trolox and were purchased from Sigma-Aldrich (St. Louis, MA, USA).
The prebiotic assessment was performed through microorganisms’ fermentation using adequate media Muller–Hinton (MH) and de Man–Rogosa–Sharp (MRS) were acquired from Biokar (Allonne, France), while cysteine used as a supplement for MRS was obtained from Merck (Darmstadt, Germany). The positive control used FOS, which was purchased from Orafti (Oreye, Belgium).

Bisphenol S Adsorption on Humic Acid
