Stomatal Morphology Visualization Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
To observe the stomatal morphology, we used a published protocol [30 (link)]. To remove any debris, leaves were acetylated in 80% ethanol for two to three min. The tiny sections of leaf were prepared using s-cutting, and after that platinum was used to fix the abaxial and adaxial surfaces and sputtered using Leica Mikrosystem GmbH (ACE600) for 25 min, and finally examined under a SEM (Thermo Scientific, Verios G4 UC, Waltham, MA, USA).

Leaf Surface Imaging by SEM

Check if the same lab product or an alternative is used in the 5 most similar protocols
For this purpose, a previously published protocol with slight modification was followed [82 (link)]. The dried leaves were first acetylated for 2 to 3 min in 80% ethanol to eliminate any debris. The s-cutting was used to prepare tiny sections of leaf, then both abaxial and adaxial surfaces were fixed on the stub, sputtered with platinum for 25 min using Leica Mikrosystem GmbH, (ACE600), and finally studied with a scanning electron microscope (Thermo Scientific, Model: verios G4 UC) present in Hainan University, Haikou China.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
