Detecting Transplanted Cells in Vivo

To locate the transplanted cells
in vivo, the expression of human ALB in liver and lung tissues of rats was detected by immunocytochemistry. The liver tissue from the 1
st and 7
th days after cell transplantation as well as the lung tissue from the first day after cell transplantation were collected to prepare paraffin sections. After being dewaxed and blocked with 5% BSA, the tissue sections were incubated with a human ALB antibody (1:300, ab137885; Abcam) in a wet box at 4°C overnight. The next day, the sections were washed with PBS, and then incubated with NL557-conjugated donkey anti-mouse IgG secondary antibody (NL007; R&D). Finally, sections were observed with an 80i Eclipse fluorescence microscope (Nikon).

Hepatic Differentiation Marker Expression

After extensive wash with PBS, the expressions of the ALB and AFP proteins in the cells was observed with a Ti-S Eclipse microscope (Nikon, Tokyo, Japan).
