Immature human MoDCs (1 × 10⁶/ml) and murine BMDCs (2 × 10⁶/ml) were treated with Mn²⁺ for 1 day. The supernatants were harvested for ELISA. Human and murine IFN-β levels were measured by Human IFN-beta Quantikine ELISA Kit (DIFNB0, R&D) and Mouse IFN-beta Quantikine ELISA Kit (MIFNB0, R&D). Interferon Beta Secretion Assay

Cells were seeded at a density of 10,000 cells per well in a 96-well
plate. Mouse IFNβ (PBL Assay Science) was then added. After 6 h of
incubation at 37 °C, the supernatant was aspirated from the wells to
remove the Mouse IFNβ. The wells were then gently washed once with
medium. Fresh warm medium was then replaced in all the wells. After 72 h of
incubation at 37 °C, the supernatant was collected, and the concentration
of IFNβ was determined using the VeriKine Mouse Interferon Beta ELISA Kit
(PBL Assay Science) or Mouse IFN-beta Quantikine ELISA Kit (R&D
Systems).

Serum Cytokine Profiling in Mice

The Apt-dimer (10 nmol) was injected into BALB/c mice via the tail vein. Whole blood was collected 4 hours after administering the injection, and serum samples were prepared by centrifugation at 1700g for 30 min. Concentrations of IL-6 and IFN-β in the serum samples were determined by using the Mouse IL-6 Quantikine ELISA Kit (M6000B, R&D Systems) and Mouse IFN-beta Quantikine ELISA Kit (MIFNB0, R&D Systems), respectively. The assay was performed according to the manufacture’s protocols. Serum samples prepared after 1.5 hours (for IFN-β) or 4 hours (for IL-6) from the single intraperitoneal injection (100 μg) of lipopolysaccharide (128-05171, Fujifilm) were used as positive controls.
